Objective:To screen the extraction technology?the filming formula of ZhanJinHuoXue film spraying agent,to investigate the dissolution and film properties of the filmed material in the alcohol-based extract of costly Chinese medicine as Musk,Bezoar,Sanguis Draxonis and so on;To optimize the preparation technology of the furacilin solution and to screen the filming formula of nitrofural film spraying agent;Investigate the quality of ginseng extract in the market;To prepare the ginseng extract anti-age serums with stable quality,and to establish its specific chromatogram of percutaneous penetration.Methods:(1)The alcohol abstraction,stream distillation-reflux extraction and percolation extract technologies were respectively used for extraction,of dracorhodin,ferulic acid and essential oil;(2)The micro image was used with the film forming properties as the index in the filming experiments with polyvinylpyrrolidone(PVP-K30)and Hydroxypropyl Methyl Cellulose(HPMC)and confirm the dosage and the proportion of them;(3)According to the solubility of the filming materials,select PVP,PVA,Methyl cellulose(MC),etc,and investigate their dissolved situation and film-forming properties in ethanol extracting solution of the different expensive Chinese Medicine by using microscope and taking pictures;(4)An initial uniform velocity method was used with the content of furacilin as the index to examine the stability of furacilin solution;(5)The micro image was used with the film forming properties and the content of furacilin as the index in the filming experiments with PVA-1788 and HPMC and confirm the dosage and the proportion of them,to optimize the filming formula of nitrofural film spraying agent;(6)The appearance of color and the content of the saponins of ginseng extract were taken as indexes to compare the quality of different ginseng extract;(7)The stability of ginseng anti-age serums was studied through the experiments of heat-resistant test,cold-resistant test observing test under room temperature;(8)The fingerprint of transdermal permeation of ginseng anti-age serums was studied through the way combine fingerprint technology with in-vitro skin permeation study.Results:(1)In the case of same menstruum dosage,the content of dracorhodin is the hightest by stream distillation-reflux extraction,followed by the alcohol abstraction,and lowest by percolation extract,the content of ferulic acid is the hightest by the alcohol abstraction,and the content of essential oil is the hightest by percolation extract,followed by the alcohol ion and its compositions of essential oil is the most complete.Therefore,the optimal technique of ZhanJinHuoXue film spraying agent was alcohol abstraction;(2)Using 5%PVP-K30 and 0.4%HPMC would quickly form a layer of uniform,density film in the ethanol extraction solution of ZhanJinHuoXue prescription;(3)The film properties of PVP in the 50%alcohol extract of Musk,Bezoar,Sanguis Draxonis were very fine;PVP was the first choice for the Chinese medicine film spraying agent;PVA-1788 in the 50%alcohol extract of Musk and Sanguis Draxonis would form a better membrane structures respectively,while form a poor membrane structures in the extract of Bezoar;MC in the 50%alcohol extract of Musk,Bezoar and Sanguis Draxonis form a poor membrane structures respectively;(4)Nitrofurazone 0.2 g,NaCl 8.5 g,boric acid 0.5 g and distilled water are maked into Nitroflural Solution with volum as 1000 mL,and define its storage validity is 1.5 years by Initial average rate methods;(5)Using 1%PVA-1788 and 0.6%HPMC would quickly form a layer of uniform,density film in the Nitroflural Solution;(6)There are many differences among ginseng extract in different company,even in the same company products have a big differences in different batches;(7)The ginseng anti-age serums is simple in preparation technology,stable in quality and suitable;(8)Establishing fingerprint spectrum system of ginseng anti-age serums in-vitro skin permeation study.5 characteristic peaks were detected in all percutaneous penetration samples in 230 nm with peak 1(ginsenoside Rgl),peak 2(ginsenoside Re),the peak 4(ginsenoside Rb1)identified.Conclusion:(1)The micro image was used with the film forming properties as the index in the filming experiments,to screen the varieties and dosages of film materials.This method can be widely applied to screen film materials for the film spraying agent.(2)This study use percutaneous penetration specific chromatogram analysis and principal component determination to explore the percutaneous permeation characteristics of ginseng anti-age serums.This method can be widely applied to study the permeability in vitro of cosmetics with Chinese herb.
