NMR Metabonomics Study Of Serum On The Model Of Abnormal Sapra Disease Carring Ulcerative Colitis

Objective:According to the theory of Uyghur traditional medicine,Abnormal Sapra syndrome rat model was established,explore the relationship between Abnormal Sapra syndrome model and Ulcerative colitis which the process of the Pathophysiological background and function.Methods:(1)According to the theory of Uygur medicine,model group rats was used the dry hot orally administered,dry hot water,under hot dry environment,stimulated with chronics and interrupted electric foot shocks and other chronic stimulation to establish abnormal spara syndrome rat model.According to the criteria of requirements,selected the developed to the characteristics of abnormal spara in rats.(2)After preparation of model of Abnormal Sapra disease carring Ulcerative colitis were induced by "DNCB+Acetic acid".Observed animal’s behavior change,compared with histopathological and ultrastructural.(3)Comparison of the behavior,Pathological and electron microscope results occurred Morphological changes each of rats,(4)A 1H NMR based metabonomic analysis was performed on these serum samples.The influential biomarkers accounting for the metabolic signature perturbation were discovered.Results:(l)Successful abnormal Sapra syndrome rat model and abnormal Sapra disease carring Ulcerative colitis was established,and the establishment of quantitative score standard.(2)Compared with the normal group,abnormal Sapra syndrome rat model underweight rats,drinking less,the difference was statistically significant(P<0.05);compared with UC control group,abnormal Sapra disease carring Ulcerative colitis weight were light,but eating much food,the difference was statistically significant(P<0.05);(3)Pathological hand electron microscope results occurred Morphological changes on abnormal Sapra syndrome rat model.compared with UC control group,abnormal Sapra disease carring Ulcerative colitis in pathology and ultrastructural is more serious;(4)compared with the normal group,abnormal Sapra syndrome rat model,abnormal Sapra disease carring Ulcerative colitis and UC control group rats serum metabolic components were different(P<0.05);compared with abnormal Sapra syndrome rat model,abnormal Sapra disease earring Ulcerative colitis and UC control group serum metabolome there were differences(P<0.05);compared with UC control rats,abnormal Sapra disease carring Ulcerative colitis serum metabolic were no differences(P>0.05);Conclusion:(1)There are morphological abnormalities and inflammatory reaction in rat colon tissue Abnormal Sapra disease rat model.(2)Abnormal Abnormal Sapra syndrome rat model had abnormal carbohydrate,lipid,protein and energy metabolism.(3)Abnormal Sapra disease carring Ulcerative colitis and UC control group rats serum metabolic component does not have the difference,but the occurrence of UC accompanied by metabolic changes.
