Effect Of Sijunzi Decoction On The Biological Characteristics Of Side Population Cells In Human Gastric Cancer Cell Line SGC-7901 In Vivo

PartⅠIsolation of side Population cells in human gastric cancer cell line SGC-7901AIM:Sorting SP cels in gastric carcinoma cell lines SGC-7901 for the next step of the experiment.Methods:Human gastric cancer cell line SGC-7901 SP cells were conventionally cultured,SP and NSP cells were sorted by Flow cytometry when the cells were in good condition.Results:The proportion of SP cells was about 1.9%.Conclusion:There were a certain number of SP cells in gastric cancer cell line SGC-7901 and could be sorted out by flow cytometry.SP cells and NSP cells were in good condition.PartⅡEffect of Sijunzi decoction on the biological characteristics of side population cells in human gastric cancer cell line SGC-7901 in vivoAIM:To investigate the antitumor effect of Sijunzi decoction on side population(SP)and non-side population(NSP)of gastric cancer cell line SGC-7901 in tumor-bearing nude mice.Methods:Establish the model of tumor-bearing mice and drug intervention group mice were given a gavage of Sijunzi decoction.All mice were killed after 30 days and kinetics of tumor formation and tumor growth were measured,tumor growth inhibition rate(IR)was calculated,apoptosis was detected by TUNEL method.Results:The mean tumor volumes of SP-Drug group,NSP-Drug group and each own model group were1348.40±103.98mm~3、628.99±98.87mm~3、987.44±84.84mm~3;2202.87±67.80mm~3、3307.76±491.44mm~3、2652.17±225.01 mm~3(P<0.05),Tumor inhibition rate of SP-drug group、NSP-drug group and unsorted-drug group were54.69%,60.19%and 56.85%,which were significantly different from the model group(P<0.05).There was also a difference between SP group and the NSP group(P<0.05).TUNEL results showed that there were more apoptotic cells in the drug groups.The apoptosis rate of SP group、NSP group and unsorted-drug group were 20.21%、16.53%and 17.34%,which were Obviously higher than model groups(P<0.05).Conclusion:Sijunzi decoction could inhibit the growth of human gastric cancer SGC-7901 SP cells in nude mice and Sijunzi decoction could induce tumor cell apoptosis.
