| Objective:The aim of this study was to explore questions as followed:1.What are the effects of EMs on HUVECs under PA?2.To investigate the mechanism of the effects that EMs on HUVECs under PA.Methods:First we get the cells from the umbilical vein of the neonate.And random allocate the cell into control group,palmitic acid group,EMI group and EM2 group.Through the technology of MTT,the cells viability under palmitic acid was inspected.In turn use the colorimetric analysis,the spectrophotometry,and enzyme-linked immunosorbent detect the levels of NO,NOS,iNOS and ET-1.Real-time PCR measure the mRNA expression of eNOS and ET-1.The contents of NO were determined in naloxone with EMs group.Result:Compared to the normal group,the cell survival rate was decreased in the palimitic acid group.The contents of NO and NOS was declined obviously,and the density of ET-1 was ascend.The expression of eNOSmRNA was cut down and the expression of ET-1 was go up significantly(P<0.05).In addition,EMs can antagonize the damage of the palimitic acid(P<0.05).Compared to the EMs groups,the level of NO was significantly decreased in naloxone with EMs groups(P<0.05).Conclusion:Palmitic acid impaired the function of HUVECs,and EMs could protect against the dysfunction via activate the μ-opioid receptor. |
