Preliminary Study On Preparation?Quality Standards And Absorption Mechanism Of Wuzi Dry Suspension

Objective:To research the technology of preparation,establish the quality standards of Wuzi dry suspension and preliminary exploration of Wuzi dry suspension and Wuzi granules absorption mechanism.Methods:(1)Use the single factor combined with central composite design-response surface methodology method,determine the contents of hyperoside,schisandrin and extract yield,an overall desirability was obtained as the index,select the best radix Cuscuta chinensis Lam.and Schisandra chinensis(Turcz.)Baill.et.medical materials to sure the aqueous alcohol extraction technology parameter;(2)Use the single factor combined with central composite design-response surface methodology method,determine the contents of total polysaccharides as the index,select the best radix Lycium barbarumL.and Rosa laevigata Michx.et.medical materials to sure the water extraction technology parameter;(3)Use the single factor combined with central composite design-response surface methodology method,determine the contents of sedimentation volume rations?electric potential?formability?absorption of moisture?viscosity?angle of repose as the index,select the best moulding technology of Wuzi dry suspension;(4)Qualitative test with thin-layer chromatography was performed on Wuzi dry suspension,with all species of medical materials,use the method of HPLC to measure the contents of hyperoside to make out the quality standard;(5)Using hyperoside as the marker compound,in situ perfusion model of rats were established to research the absorption characteristics of Wuzi dry suspension and granules.Results:(1)The optimum ethanol extraction procedure was addingl2 times amount of 55%ethanol,extracted two times,each time 30min;(2)The optimum water extraction procedure was adding 12 times amount of water extracted two times,each time 165min;(3)The dry suspensions' moulde technology was using 18%mixed excipients(8%xanthan gum-2%sodium alginate-8%sodium carboxyl methyl cellulose)and 88%ethanol as bond to make granules;(4)Established the qualitative differentiation with the method of TLC of Lycium barbarumL and Schisandra chinensis(Turcz.)Baill.,and the content of hyperoside was detected by HPLC,the mobile phases:methanol-0.5%HCOOH(39:61)detect wavelength:360nm column temperature:30?,flow rate:1.0mL/min;(5)Preliminary analysis showed that the absorption of hyperoside may be the first order kinetics.Conclusions:The extraction technology was stable,feasible.The quality standards were reliable and accurate,and can effectively control the quality of the preparations.Intestinal absorption kinetics of Wuzi dry suspension prepared for the bioavailability of TCM formulas.
