ObjectiveHepatitis B virus(HBV)is the most common cause of acute and chronic liver disease worldwide,and it has been a major public health problem.HBV-infected adults can develop into acute HBV infection or chronic hepatitis B which may result in liver cirrhosis or hepatocellular carcinoma.But the reasons of different progressions of the HBV infection were not clear,Substantial evidence showed that the chronic hepatitis B was associated with host immunological factors,sex,age and genetic factors.Recent studies explored the role of Mannan binding lectin 2(MBL)and Interleukin-10(IL-10)playing in progression of the HBV infection.This study used TagSNPs and hot snps selecting principles to investigate the possible association between MBL2 and IL-10 genes polymorphisms and susceptibility to chronic hepatitis B in Chinese Han population.MethodsThe case-control population contained 1297 unrelated adult Han Chinese,who were recruited from the outpatient clinics and hospitalization wards at the First Affiliated Hospital of Anhui Medical University and the Second Affiliated Hospital of Anhui Medical University。Of these,996 chronic HBV infection patients(693 males and 303 females)were recruited at Infectious Disease department as cases and 301 acute HBV infection individuals as controls(135 females and 166 males),and Hepatitis B markers of all patients were tested。DNA was prepared from peripheral blood leucocytes by standard methods.Multiplex Snapshot Technique methods were used in MBL2 and IL-10 genotyping.Statistical analysis was done by using SPSS 13.0,Haploview and PLINK3.1 software.Test level was 0.05(a=0.05)and Bonferoni correction was used to adjust P value.RESULTS1.996 chronic HBV infection patients(693 males and 303 females)were recruited at Infectious Disease department as cases and 301 acute HBV infection individuals as controls(135 females and 166 males).The age was no significantly different between the chronic HBV infection cases and the self-limited controls in all samples,but sex was significantly different between the chronic HBV infection cases and the self-limited controls(X2=21.516 and P<0.001).2.The 3 TagSNPs(rs2120131,rs4935047 and rs7095891)in MBL2 and two snps(rs3024490 and rs3021094)in IL-10 were conformed to the HWE in each group.3.We found none of the TagSNPs(rs2120131,rs4935047 and rs7095891)of MBL2 had significantly different between the chronic HBV infection cases and the self-limited controls in all samples,females subgroup and males subgroup in allele analysis,different genetic models analysis or haplotype analysis(adjusted P>0.05).4.The SNPs rs3021094 of IL-10 was significantly different between the chronic HBV infection cases and the self-limited controls in all samples,females subgroup and males subgroup in allele analysis,different genetic models analysis(adjusted P>0.05).5.The rs3024490 allele frequencies were significant difference in all sample(adjusted P<0.001,OR=1.444,95%C=1.185-1.761),male sample(adjusted P=0.008,OR= 1.472,95%CI=1.131-1.915)and female sample(adjusted P=0.030,OR=1.462,95%CI=1.076-1.987)6.The rs3024490 in all samples were significant different between chronic HBV infection cases and the self-limited controls in additive genetic model(adjusted P=0.002),dominant genetic model(adjusted P=0.002),and recessive genetic model(adjusted P=0.026).The rs3024490 in male samples were significant different between chronic HBV infection cases and the self-limited controls in additive genetic model(adjusted P=0.030)and dominant genetic model(adjusted P=0.022),except in recessive genetic model(adjusted P=0.070).The rs3024490 in female samples were significant different between chronic HBV infection cases and the self-limited controls in dominant genetic model(adjusted P=0.022).7.In haplotype analysis,we only found haplotype AG of IL-10(rs3021094|rs3024490)was significant different between chronic HBV infection cases and the self-limited controls(in all samples:adjusted P =0.045,OR=1.390.In male samples:adjusted P =0.007 OR=1.450.In male samples:adjusted P =0.045,OR=1.360.)Conclusions1.No association between three new snps(rs2120131,rs4935047 and rs7095891)within MBL2 polymorphisms and susceptibility to chronic hepatitis B in Chinese Han population.2.The rs3024490 G allele within IL-10 was a risk factor of susceptibility to chronic hepatitis B in Chinese Han population.
