| ObjectiveTo observe the resveratrol and the Jiedu Sangen Decoction(Polygonum cuspidatum root,root Bennet,Actinidiae)impact on CT-26 colon cancer cell invasiveness and research CAFs in Sirt1-NF-κB related effects on explore detoxification the Jiedu Sangen Decoction in the carcinoma microenvironment and provide a theoretical basis for further clinical and research laboratory.Method1)The BALB/C mice were randomly divided into normal control group,model group and Jiedu Sangen Decoction in 3 group.Molding for 7 days in Successful,to gastric lavage the normal control group and model group for 10 days with physiological saline,Jiedu Sangen Decoction was gastric lavage for 10 days,blood grouping serum was separated spare;2)CT-26 cells to recover,passaged cells collected logarithmic growth phase,transwell law and observe the effects of resveratrol on 3 serum CT-26 cell migration force;3)carcinoma-associated fibroblasts(CAFs)to establish:CT-26 supernatant was collected,and the ratio of 1:1 with fresh medium configuration conditioned mediun;collection logarithmic phase of mouse fibroblast L929 cells,with condition medium for 72 hours to establish CAFs model;4)After the serum and 24 hours after the intervention of resveratrol,western blot detection interfere with the normal mouse serum containing fibroblasts,NF-κB pathway and Sirtl expression levels;5)Immunofluorescence analysis of CAFs in NF-κB and the expression of Sirtl pathway and the role and position.Result1)Transwell results:Compared to other groups,Jiedu Sangen Decoction and resveratrol group can reduce the number of CT-26 colon cancer cells,the difference was significant(P<0.01);2)Western Blot results showed that:the expression of 24 hours of Jiedu Sangen Decoction and resveratrol group CAFs was compared with the other groups that was increased Sirtl significantly.Jiedu Sangen Decoction groups and resveratrol group than other groups to express signifieant phosphorylation of P65 reduced significantly increased the phosphorylation of IKBa,phosphorylation IKKap expression.There was statistically significant(P<0.05);3)Immunofluorescence showed that Jiedu Sangen Decoction and resveratrol can enhance Sirtl’ fluorescence than other groups,and interact with NF-κB in the nucleuS.Conclusion1)Jiedu Sangen Decoction and resveratrol can inhibit migration and invasion of movement CT-26 colon cancer cells;2)Jiedu Sangen Decoction can enhance the expression of Sirt1 CAFs and activ of the NF-κB pathway CAFs inhibition. |
