Background:Osteosarcoma is a common primary malignant bone tissue in the bone primary bone tumors,the incidence of osteosarcoma after plasma cell myeloma ranks second,accounting for primary bone 20%of tumors,and 75%of patients 10 to 30 years old,a high degree of malignancy,seriously affecting the health of young adults.From the beginning of the 1970s,began to be used chemotherapy drugs in patients with osteosarcoma,and achieved remarkable results.To the late 1970s and early 1980s and gradually formed a concept of neoadjuvant chemotherapy.Rosen proposed neoadjuvant chemotherapy concept in 1982,which has become the standard mode of treatment of osteosarcoma.But there are still many difficulties in the clinical treatment,to explore new,effective chemotherapy treatment for osteosarcoma will help to improve the survival time of patients with osteosarcoma.Matrine is an active substance extracted from traditional Chinese medicine Sophora of dried roots belonging tetracyclic quinolizidine class,in recent years studies have found that many of its in vitro inhibition of tumor cell proliferation,induction of apoptosis,inhibition of tumor cell adhesion and metastasis,and drug resistance and reversal of inhibition of tumor cells and so on,but also in clinical flavescens and matrine on leukemia,liver cancer and stomach cancer treatment is effective coverage.Objective:To study the cytotoxicity effect and mechanism of matrine on human osteosarcoma cell line MG-63,which could provide experimental evidence to prospects for the treatment of osteosarcoma.Mrthods:Observed the morphology of human osteosarcoma cell under inverted microscope.The proliferation effect of MG-63 cell line cultured with matrine in different concentrations and variable durations,at the same time was detected by MTT assay.Alteration of cell cycle of different concentration treating and control groups at the second day was analyzed by Flow cyctometry(FCM).Results:Compared the control group with 0.4,0.8,1.2,1.6,2.0mg/ml treatment groups,the cell of matrine-treated groups showed reduction of volume and karyoplasmic ratio.Matrine can inhibit proliferation of MG-63 cells.The effect of proliferation inhibition was more prominent with the treated time prolonging and the drug concentration increasing,which showed a time-dependent and dose-dependent manner.Increased G1 phase rate,decreased S phase rate were showed by FCM after matrine-treated MG-63 cells.Conclusion:Matrine has cytotoxicity against human osteosarcoma cell line MG-63.The mechanism includes that matrine can effectively inhibit the proliferation of MG-63 cell line,arrest the cell cycle at G1 phase and induce the apoptosis of MG-63 cell line.The effect of matrine displays a time-dependent and dose-dependent manner.
