Objective:Berberine were given for intervention after the 3-month.Test abnormal GSK3p phosphorylation and activity in Polycystic ovarysyndrome(PCOS)patients ’ ovarian granulosa cells,and the impact on ovarian granulosa cells insulin resistance and androgen excess.Through the experiment to prove the abnormal GSK3β phosphorylation and activity was a causati,vefactor in PCOS,and the effect of berberine on ovarian granulosa cells insulin resistance and androgen excess is jointly controlled by GSK3 in women with PCOS.Design and Methods:All of patiens have BMI≧23kg/m2,18<age<35.The ovarian granulosa cellsfrom 5 non-PCOS patiens and 10 of PCOS patients,which 5 PCOS patiens had eated 3-month berberine,5 PCOS patients had no intervention.the diagnosis of PCOS presupposes the presence of at least two out of the three following features:anovulation(ANOV),clinical and/or biochemical hyperandrogenism(HA)and polycystic ovaries on ultrasonography(PCO),with other androgen excess disorders excluded.Subsequently,additional milder phenotypes of the syndrome have been included inPCOS;still,phenotypes with biochemical hyperandrogenemia and chronic anovulation appear to be the most severe in terms of androgen levels,insulin resistance and obesity.PCOS standard Ovarian granulosa cells were collected after IVF/ICSI-ET andwere cultured in vitro for 48h.The following techniques were used:by radioimmunoassay ascertain by measuring the level of testosterone,estradiol,Progesterone;utilizingglucose assay kit(HK Method)to test cell culture medium residual sugar;exploring the 2-NBDG to assessment cell glucose uptake;using GSK-3pactivity kit to detect the activity of GSK-3P;and were analyzed GSK3β,P-GSK3β(ser9,tyr216)by western-blot.Results:Androgen secretion within ovarian granulosa cells from non-PCOS and berberine,which compaired with PCOS patients,was decreased separately(p<0.05),non-PCOS had a higher progesterone level than PCOS(P<0.01),so did berberine(P<0.05).But all group had not change significantly of estradiol(P>0.05);culture liquid residual sugarofnon-PCOS and berberine is lower than PCOS(P<0.01).2-NBDG showed glucose uptake berberine is increased(P<0.01),non-PCOS is invreased(P<0.01)too;GSK3p activityof berberine is lower than PCOS(P<0.05).non-PCOS have the same tend(P<0.01).western-blot showed that P-GSK3p(ser9)was decreased expression in ovarian granulosacells from PCOS patients(P<0.01),and high expression of P-GSK3β(tyr216)(P<0.05),but berberine have a contrary tend.Conclusions:1.Ovarian of PCOS patients have glucose metabolism disorders and higher androgen secretion.Berberine can reduce androgen secretion and improve abnormal glucose metabolism.2.Berberine can be regarded as GSK3pinhibitors,which inhibited GSK3p activity and regulated the abnormal GSK3p phosphorylation in ovarian granulosa cells from PCOS patients.3.GSK3pis a partial cause of PCOS patients responsible for impaired glucose uptakeand androgen excess in ovarian granulosa cells from PCOS patients.Berberine play a key role in treating PCOS.
