| Objective:By acupuncturing“Baihui through Qubin”,the expression of GSK-3β in rat brain tissue of acute cerebral hemorrhage can be observed,in order to study the relationship between the treatment and the cerebral protection mechanism during acute cerebral hemorrhage.Methods:There are 4 groups in the experiment:blank group,model group,acupuncture group and inhibitor group,while in the last 3 groups,each group is distributed into four subgroups again:they are 1 day subgroup,3 days subgroup,7 days subgroup and 14 days subgroup.Now there are 13 sub groups in total.156 male Wistar rats are randomly distributed into the above 13 groups,each has 12 rats.The rats in the blank group have no treatment,while the cerebral hemorrhage models should be established on the rats in other three groups.The neurological function score is needed to select eligible rats for future research.After different groups of acute cerebral hemorrhage rats have been accepted corresponding treatments in different period of times,it is the time to get rat brain tissue and to observe the injured rat brain tissue under microscope.In order to observe the further mechanism of acute cerebral hemorrhage,Western blot method and Immunohistochemical method,which are used to study the amount of positive cells of GSK-3β and the condition of the expression of GSK-3β protein,are also needed in the experiment.In each group,6 rats are selected to do Western blot method,while the other 6 rats are selected to do Immunohistochemical method.Results:1.Obvious brain tissue damage of model group rats can be seen under the microscope,which are accompanied with liquefaction,dropsy and necrosis,etc,which has changed extremely in the third day and gradually relieved after this time point.However,compared with the model group,the brain tissue damage of acupuncture group and inhibitor group are more abirritant in each corresponding time point.2.The result of GSK-3β Western blot method:The expression of the amount of GSK-3β protein of model group,inhibitor group and acupuncture group is generally in accordance with the observation of the brain tissue damage under the microscope in different period of times.The expression of GSK-3β protein of model group,inhibitor group and acupuncture group reduced in the third day,and in the seventh day,the expression of GSK-3βprotein of the above three groups is more than before,while in the fourteenth day,the expression of GSK-3β protein of the above three groups is less than the expression in the seventh day.The effect of acupuncture group is much better than model group,which shows the statistical significance(P<0.05).The effect of inhibitor group is also better than model group,which shows the statistical significance(P<0.05).However,there is no statistical significance between inhibitor group and acupuncture group(P>0.05).3.The result of GSK-3β immunohistochemical method:The results between immunohistochemical method and Western blot method are similar.The expression of GSK-3p protein of model group,inhibitor group and acupuncture group reduced in the third day,and in the seventh day,the expression of GSK-3β protein of the above three groups is more than before,while in the fourteenth day,the expression of GSK-3β protein of the above three groups is less than the expression in the seventh day.The effect of acupuncture group is much better than model group,which shows the statistical significance(P<0.05).The effect of inhibitor group is also better than model group,which shows the statistical significance(P<0.05).However,there is no statistical significance between inhibitor group and acupuncture group(P>0.05).Conclusions:1.By using the treatment of acupuncturing "Baihui through Qubin",several kinds of pathological injuries of acute cerebral hemorrhage can be controlled,and the treatment can protect the nervous system.2.By using the treatment of acupuncturing Baihui through Qubin",the expression of GSK-3β in corresponding signal pathway,which can boost the regeneration of nerve cells,can be inhibited. |
