| Objective The purpose of this study is to construct epithelial ovarian cancer cisplatin-resistant cell lines TOV112D/DDP,and to detect the expression of human epidermal growth factor receptor 2(Her2)and the activation of signal transducers and transcription activators 3(STAT-3)in related signaling pathways of the cisplatin-resistant cancer cells by the method of western blot and immunofluorescence cytochemistry,and to discuss the association between the resistance of epithelial ovarian cancer and the expression of gene Her2 and its activation of related signaling pathways,then to provide the theoretical basis for the clinical treatment of drug-resistant ovarian cancer.Method ① The epithelial ovarian cancer cisplatin-resistant cell lines(TOV112D/DDP)had been constructed by the micro-incremental method.②The evaluation of drug resistance of TOV112D/DDP had been detected the half maximal inhibitory concentration of a substance(IC50)of TOV112D and TOV112D/DDP by MTT testing,evaluated the resistance of TOV112D/DDP.③ The Her2 expression of TOV112D/DDP:had been evaluated by the methods of western blot and immunofluorescence cytochemical techniques.④ The STAT3 expression and action of TOV112D/DDP had been dectected by the methods of western blot and immunofluorescence cytochemical techniques.Results ① The epithelial ovarian cancer cisplatin-resistant cell lines(TOV112D/DDP)had been successfully constructed,the MTT assay result showed that the cisplatin resistance coefficient of TOV112D/DDP had been 4.1429.②Compared with TOV112D,the Her2 expression of cisplatin-resistant cells TOV112D/DDP had been significantly up-regulation,and the result of the confocal laser scanning microscopy had been showed the consistent results above.③The STAT3 expression of cisplatin.resistant cells TOV112D/DDP of western blot analysis had been significantly upregulation camparing with TOV112D.and the results of confocal laser scanning micro microscope had found that STAT3 of cisplatin-resistant cells TOV112D/DDP had been significantly upregulation camparing with TOV112D,and cisplatin-resistant cell nucleus had been particularly significant upregulation of STAT3.④ The results of confocal laser seanning mieroscopy had showed that p-STAT3 had been mainly expressed in nucleus of TOV112D and TOV112D/DDP,the results of both western blot and confocal scanning microscopy had showed that the TOV112D/DDP cells expression of p-STAT3 had been significantly increased compared with TOV112D.Conclusion ① Successfully constructed the epithelial ovarian cancer cisplatin-resistant cell lines of(TOV112D/DP).②The Her2 expression of cisplatin-resistant cells TOV112D/DDP had been significantly increased compared with TOV112D.③ The STAT3 expression of cisplatin-resistant cells TOV112D/DDP had been significantly increased compared with TOV112D.④ Compared with TOV112D cells,Cisplatin-resistant cells TOV112D/DDP activation levels of STAT3 i.e.p-STAT3 expression level had been significantly increased. |
