| Rheumatoid arthritis(RA)is a systemic autoimmune disease.It is a chronic progressive symmetric arthritis.It can lead to joint destruction,deformity and dysfunction,and even disability.The etiology of this disease is still unclear,generally think genetic,infection,environmental factors is the main reason of this disease.The root tuber of Thladiantha dubia Bunge is the ripe root of cucurbitaceous plant Thladiantha dubia.Long-term application of RTD decoction among the folk has proved that it has strong anti effect.RTD has been affirmed with a clear effect of resistance to rheumatoid arthritis and has less noxious side effect for a long-term practical application.It mainly contains triterpenoid saponins.Experimental studies had show that saponins of Thladiantha dubia root(STDR)has the therapeutic action on the model of adjuvant arthritis(AA)ratsPart 1 The Therapeutic Activity Saponins of Thladiantha dubia Root in Rats Model of Adjuvant ArthritisObjective:To observe the effect of STDR on the pathological change of the secondary hind paws,the pathological change of the synovial membrane and the spleen index in rats model with Adjuvant Arthritis.To study the therapeutic effect in rats model of AA.Methods:1.Take 10 of 80 male rats into normol control group random.The others rats were induced AA by inject FCA into the right hindlimb.The rats which induced AA divided in six groups of ten each.The normol control group,model group,STDR H(160 mg/kg/d),STDR M(80 mg/kg/d),STDR L(40 mg/kg/d)groups and Tripterygium Glycoside 12 mg/kg/d group.STDR H(160 mg/kg/d),STDR M(80 mg/kg/d),STDR L(40 mg/kg/d)groups and Tripterygium Glycoside 12 mg/kg/d group.The STDR groups and Tripterygium Glycoside group rats gavage administration for 21 days.2.The secondary hind paw swelling degree and the arthritis index of rats was observed before medication and the 5th days,9th days,13 th days,17 th days,21 st days after medication.3.The pathology of secondary hind paw and knee joint synovial membrane of each group was observed by hematoxylin-eosin staining after medication.4.Weigh the rats spleens of each group,calculate the spleen index of each group.Results:1.The secondary hind paw swelling degree of AA rats in NC groups were lower than those in model group(P<0.01).The secondary hind paw swelling degree of AA rats in STDR H,STDR M and STDR L groups were lower than those in model group after gavage administration for 5,9 and 13 days(P<0.05).The secondary hind paw degree of AA rats in STDR groups were lower than those in model group obviously after gavage administration for 21 days(P<0.01).The AI of AA rats in NC groups were lower than those in model group(P<0.01).The AI of AA rats in STDR H and TG groups were lower than those in model group after gavage administration for 9 days(P<0.05),The AI of AA rats in STDR M and STDR L groups were lower than those in model group after gavage administration for 13 days(P<0.05).The AI of AA rats in each group were lower than those in model group obviously after gavage administration for 21 days(P<0.01),。2.The muscular tissue of secondary hind paws of the NC group was completion.The muscular tissue of secondary hind paws of the model group was necroses,inflammatory infiltration and fibrosis was severe,inflammatory infiltration was inhibited in STDR H,STDR M and TG groups.The fibrosis was inhibited in STDR H and TG groups.3.The knee joint synovial membrane of NC group was the whole connective tissue,the synovial membrane of model group has the inflammatory infiltration and hyperplasia inflammatory,infiltration and hyperplasia of knee joint synovial membrane of each STDR group was inhibited.3.Compare with the NC group,the SI of model group was increase obviously.SI of AA rats in STDR M,STDR H groups were lower than those in model group(P<0.05 or P<0.01).Conclusions: The STDR has the inhibit effect on inflammatory response of the secondary hind paw,inflammatory response of the knee joint synovial membrane and spleen swelling in AA rats.Part 2 The Effect on Expression of TNF-α,IL-1β,IL-6 of Total Saponins of Thladiantha dubia Root in Rats Model of Adjuvant ArthritisObjective:To study the effect of total saponins of Thladiantha dubia root on the expression levels of TNF-α,IL-1β and IL-6 in secondary hind paws and spleens in adjuvant arthritis rats.Methods:1.The expression of TNF-α,IL-1β and IL-6 m RNA in the secondary hind paws of AA rats was determined by RT-PCR.2.The expression of TNF-α,IL-1β and IL-6 in the secondary hind paws of AA rats was determined by immunohistochemistry assay.3.The expression of TNF-α,IL-1β and IL-6 m RNA in the spleens of AA rats was determined by immunohistochemistry assay.Results:1.The expression of TNF-α,IL-1β and IL-6 m RNA in the secondary hind paws in NC group were higher than those in model group(P<0.01).The expression of TNF-α,IL-1β and IL-6 m RNA in STDR groups and TG group were lower than those in model group(P<0.05 or P<0.01).2.The expression of TNF-α in the secondary hind paws in NC group were higher than those in model group(P<0.01).The expression of TNF-α in STDR L group was lower than that in model group(P<0.05)and in STDR M,STDR H,TG groups were lower than those in model group.The expression of IL-1β in the secondary hind paws in NC group were higher than those in model group(P<0.01).The expression of IL-1β in STDR L group was lower than that in model group(P<0.05)and in STDR M,STDR H,TG groups were lower than those in model group.The expression of IL-6 in the secondary hind paws in NC group were higher than those in model group(P<0.01).The expression of IL-6 in STDR L group was lower than that in model group(P<0.05)and in STDR M,STDR H,TG groups were lower than those in model group.3.The expression of TNF-α in the spleens in NC group were higher than those in model group(P<0.01).The expression of TNF-α,IL-1β and IL-6 in STDR L group was lower than that in model group(P<0.05)and in STDR M,STDR H,TG groups were lower than those in model group.The expression of IL-1β in the spleens in NC group were higher than those in model group(P<0.01).The expression of IL-1β in STDR L group was lower than that in model group(P<0.05)and in STDR M,STDR H,TG groups were lower than those in model group.The expression of IL-6 in the spleens in NC group were higher than those in model group(P<0.01).The expression of IL-6 in STDR L group was lower than that in model group(P<0.05)and in STDR M,STDR H,TG groups were lower than those in model group.Conclusions:The STDR has Anti-inflammatory effect on AA rats.The role is closely related to the inhibition on the expression of cytokines TNF-α,IL-1β and IL-6. |
