| Objective To research the influence of high fat diet on the female rats’expression of hypothalamus GnRH mRNA,Kiss-1 mRNA,GPR54 mRNA and pituitary GnRH-R mRNA,and the intervention mechanism of Maturation NO.2 on it.Methods Choosing 30 female SD rats of 21 days old,randomly divided them into 3 groups,namely high-fat group,control group and CHM group,10 in each group.Control group was fed with standard feed.High-fat group and CHM group were fed with high-fat feed.At the same time,CHM group was gavaged with Maturation N0.2,while the others were gavaged with normal saline.When the 10 rats in the high-fat group appeared to have vaginal opening,all the rats were going to be executed to obtain the hypothalami and pituitaries.The relative expression of hypothalamus GnRH mRNA,Kiss-1 mRNA,GPR54 mRNA and pituitary GnRH-R mRNA were detected by RT-PCR.Result The relative expression of hypothalamus GnRH mRNA,Kiss-1 mRNA,GPR54 mRNA and pituitary GnRH-R mRNA:high-fat group was much higher than control group(P<0.05).CHM group was lower than high-fat group,but there was no significant difference between them(P>0.05).Conclusion Long-term high-fat diet can cause the female rats’ function of HPGA to start ahead of schedule.Maturation NO.2 has no significant inhibitory effect on the precocious puberty caused by high-fat diet. |
