Malignant tumor has been one of frequently-occuring diseases that threat the human life.Hence,the search and development of novel antitumor drugs has become a hotspot in the field of medcinal chemistry.Microbial metabolites have been proved to be a rich source of lead compounds for antitumor drugs developement.In this paper,we report the isolation,structure elucidation,and cytotoxicity evaluation of the secondary metabolites produced by Neosartorya glabra.The paper was devided into three chapters.In the first chapter,progress on the secondary metabolites of Neosartorya glabra was reviewed,and the significance of this research was also proposed.The second chapter concerned the extraction,purification and structural identification of secondary metabolites of Neosartorya glabra.Thirteen compounds were isolated from the wheat culture of Neosartorya glabra They were elucidated as neosarmin A(1)、6-hydroxy-methoxyvermistatin(2)、3’-methylbuta-dehydroisopenicillide(3)、berkeleyacetal D(4)、phialophoriol(5)、chrodrimanin A(6)、6-demethylvermistatin(7)、penicillide(8)、berkeleyacetal C(9)、chrodrimanin B(10)、vermistatin(11)、purpactin A(12)、methoxyvermistatin(13)using spectroscopic methods(UV、IR、1D-NMR、2D-NMR、MS)and chemical analysis.Among these compounds,1-4 were new compounds.In the third chapter,cytotoxicity evaluation of compounds 1-13 was reported.The cytotoxic activity for 1-13 was evaluated by the MTT method against two Breast Tumor Cells(MDA-MB-231 cells and MCF-7),Human Umbilical Vein Endothelial Cells(HUVEC),Pancreatic Tumor Cells(PANC-1).Among the compounds tested,the new compound 2 and the known compound 8 showed moderate inhibitory effects on the proliferation of PANC-1 cells,the IC50 values of which were 14.38±0.51 μM and 10.93±0.21 μM,respectively.
