Background: Gastric cancer is one of the most common malignant tumors in the world.The incidence and mortality of gastric cancer are fifth and third respectively.In China,gastric cancer is the most common malignant tumor in digestive system.The incidence and mortality of gastric cancer is the second most common in malignant tumors,only next to lung cancer.The occurrence of gastric cancer is a process of many factors involved in multiple stages.It is also the result of genetic,HP infection and environmental factors.At present,the research of gene has become a hot spot in the pathogenesis of gastric cancer.Many genes were found to be abnormal in gastric cancer,and correlated with the occurrence and progression of this malignancy.Many members of the keratin family are among them.In recent years,the relationship between cytokeratin and gastric cancer have received more and more attention.Keratin referred to as KRT or CK,is an integral part of the intermediate filaments of the cytoskeleton protein,which is composed of more than 50 members,and can be divided into Type I and Type II according to the difference of p H and isoelectric point.Keratin plays an important role in maintaining the mechanical stability and integrity of epithelial cells and tissues.Keratin is usually expressed in normal epithelial cells and tumor cells of epithelial origin,when canceration of epithelial cells or malignant epithelial tumor metastasis,tumor cells can still keep the cell of origin of keratin type.According to this characteristic,we can identify the source of unknown primary tumor by detecting keratin.In addition to normal expression in gastric tissues,more and more studies have shown that the expression of m RNA and protein in the keratin family members is abnormal in gastric cancer.Including CK20,CK7,CK18,CK19,their expression levels were closely related to tumor differentiation,lymph node metastasis,clinical stage,prognosis and so on.Keratin 80 is a member of the Keratin gene family.It belongs to type II epithelial keratin,which is located in human chromosome 12q13.13,and its encoded protein contains 452 amino acids,molecular weight of 50.5 k Da,its isoelectric point is about 5.47.At present,there are few researches on KRT80 m RNA and protein expression in China and abroad.The expression of Keratin 80 in gastric cancer has not been reported in literature.Objective: In order to investigate the expression of KRT80 in gastric carcinoma and its relationship with the clinicopathological features of this malignancy.Method: We collected 20 cases of gastric cancer tissues and corresponding paracancerous tissue specimens in the Department of gastrointestinal surgery and general surgery of Yantai Yuhuangding Hospital from March 2016 to 2016 in July.The postoperative pathology diagnosis is gastric adenocarcinoma.All patients were not treated with chemotherapy,radiotherapy and targeted therapy before operation.In this study,the expression of KRT80 protein and m RNA in gastric cancer and adjacent tissues were detected by western blot and fluorescence quantitative PCR respectively.The relationship between KRT80 protein expression and clinicopathological features of gastric cancer was analyzed.Results: The expression of KRT80 m RNA and protein were detected in 20 cases of gastric cancer tissues and corresponding adjacent tissues.The average expression level in gastric cancer tissues was significantly higher than that in adjacent tissues.The expression of KRT80 protein was not significantly correlated with age,sex,tumor differentiation,depth of invasion,tumor size and TNM stage(P<0.05),but was associated with lymph node metastasis(P<0.05).Conclusion:It was found that the expression of KRT80 m RNA and protein in gastric cancer and adjacent tissues,up-regulated expression of KRT80 in gastric carcinoma.The expression of KRT80 protein was correlated with lymph node metastasis in patients with gastric cancer.
