Expression And Significance Of Calcium-activated Chloride Channel Transmembrane Protein 16A In Colorectal Cancer And Adenomatous Polyps Patient

Objective 1.To investigate whether there are differences in the expression of TMEM16 A in adenomatous polyps,colorectal cancer and adjacent normal tissues;2.To investigate the relationship between the expression of TMEM16 A in colorectal carcinoma and its correlation with tumor location,gross type,differentiation degree,depth of invasion,tumor TNM stage,lymph node metastasis,distant metastasis,serum CEA and so on;Method 1.40 cases of colorectal cancer in The first affiliated hospital of dalian medical university were enrolled,the colorectal cancer tissues and paracancerous tissues were collected during the operation.2.40 cases of adenoma in The first affiliated hospital of dalian medical university were enrolled.3.The expression of TMEM16 A was detected by immunohistochemistry,Real-Time PCR and Western Blot in colorectal cancer tissues,paracancerous tissues and adenomatous polyps respectively.The difference of TMEM16 A expression in the different groups according to the age,degree of differentiation,depth of invasion,TNM staging,metastasis,lymph node metastasis,serum CEA level,tumor gross type were analysed.Result 1.The differences between the expression of TMEM16 A in adenomatous polyps,colorectal cancer and adjacent normal tissues: Immunohistochemical detection show that the positive rate of TMEM16 A protein was 75% in colorectal cancer tissue,while 7.5% in colorectal cancer tissue and 50% in adenoma,the difference had statistically significant between the three groups(P < 0.05).Results of Real-Time PCR and Western Blot show that the expression of TMEM16 A in colorectal cancer tissues was higher than paracancerous tissues(P < 0.05).2.The relationship between the expression of TMEM16 A in colorectal carcinoma and its correlation: Real-Time PCR and Western Blot results show that the expression level of TMEM16 A in III+IV stage group was higher than that in the I+II stage group;and the distant metastasis group was higher than without distant metastasis;the expression level in lymph node metastasis group were higher than that in the group without lymph node metastasis,the expression level in increased serum CEA group was higher than that of the normal group,these differences had statistically significant(P < 0.05),while there was no significant difference of TMEM16 A expression according to different gender,age,degree of differentiation,depth of invasion,gross type,tumor site(P > 0.05).Conclusion 1.TMEM16 A is over-expressed in colorectal carcinoma.2.High expression of TMEM16 A may be related to metastasis of colorectal cancer.3.TMEM16 A may be involved in carcinogenesis of colorectal adenomatous polyps.
