Objective:Observe the effect of Huatan-Tongyu-Jiedu Decoction(HTJD)aqueous extract on the invasion and metastasis and the expressions of associated protein of Wnt/β-catenin signal pathway on human gastric cancer cell line SGC-7901,to explore the mechanism on of Huatan-Tongyu-Jiedu Decoction to inhibition invasion and metastasis of human gastric cancer cell lines SGC-7901.Method:Treated the human gastric cancer SGC-7901 cells with different concentrations of HTJD aqueous extract(5,10,20,30,40μg/m L)respectively.Transwell assay and wound healing experiment were used to detect the effect of SGC-7901 on invasion and metastasis of human gastric cancer cell line SGC-7901.Enzyme linked immunosorbent assay was used to detect the effect of the expressions of Epithelial cadherin(E-cadherin)、Cyclooxygenase-2(COX-2)、matrix metallopro-teinase 2(MMP-2)and 44 Cluster of differentiation variants6(CD44v6)of human gastric cancer cell line SGC-7901 on HTJD;Western blotting assay was used to detect the effect of the relative expressions of Wnt gene family factor 1(Wnt1)、β-catenin、glycogen synthase kinase-3β(GSK-3β)and Adenomatous polyposis of the colon protein(Apc)of human gastric cancer cell line SGC-7901 on HTJD.Results:.1.Effect of HTJD on invasion and migration of human gastric cancer cell line SGC-7901.Compared with the control group,the number of cells penetrating through the Transwell was significantly reduced in the groups of 10μg/m L,20μg/m L,30μg/m L 、40μg/m L;wound healing distance △d reduced significantly(P<0.05 orP<0.01).2.Effects of HTJD on the expression of E-cadherin,MMP-2,COX-2 and CD44v6 in human gastric cancer SGC-7901 cellsThe expression of E-cadherin was significantly increased in SGC-7901 cells(P<0.05orP<0.01),the expression of COX-2,MMP-2and CD44v6 was significantly increased in SGC-7901 cells in the groups of 10μg/m L、20μg/m L、30μg/m L、40μg/m L(P<0.05 orP<0.01),Showing a dose-dependent in the groups of 20、30、40μg/m L.3.Effects of HTJD on the expression of associated protein of Wnt/β-catenin signal pathway on human gastric cancer cell line SGC-7901The expression of Apc and GSK-3βwas significantly increased in SGC-7901 cells(P<0.05 orP<0.01),the expression of Wnt1 and β-catenin was significantly increased in SGC-7901 cells in the groups of 10μg/m L、20μg/m L、30μg/m L、40μg/m L(P<0.05 orP<0.01),Showing a dose-dependent in the groups of 20、30、40μg/m L.Conclusion1..Huatan-Tongyu-Jiedu Decoction can significantly inhibit the invasion and migration of human gastric cancer cell SGC-7901,can be raised E-cadherin of human gastric cancer SGC-7901 cells,down-regulation of MMP-2,CD44v6 and COX-2 expression2.Huatan-Tongyu-Jiedu Decoction can inhibit the invasion and migration of human gastric cancer cell SGC-7901 and its regulation of Wnt/β-catenin signal transduction pathway-related protein expression is closely related.
