Objective:To establish a model of cardiomyopathy induced by short ventricular tachycardia;To observe the expression of TGF-β1 / Smad signaling pathway in patients with tachycardia.Methods: Twenty-six healthy New Zealand rabbits were randomly divided into A and B groups.All New Zealand rabbits were injected with 6F vascular sheath from the right jugular vein.(6 rats),group B for the experimental group(20),the right ventricular stimulation 400 times / min modeling,each group continued for 5 to 8 A room early formation of short array of ventricular tachycardia,30 sets / minute,the cumulative daily stimulation of 3 hours.The left ventricular end diastolic diameter(LVEDD)was measured in all New Zealand rabbits before and after stimulation for 1week,4 weeks,8 weeks and 12 weeks after stimulation.(LVESD)and left ventricular ejection fraction(LVEF)were measured.Four New Zealand rabbits were randomly selected and sacrificed.Myocardium specimens were collected and HE staining was performed.RT-PCR was used to detect TGF-β1,Smad2 and Smad3 Expression of Smad7 in New Zealand Rabbit Myocardium.Results: 1.Animal survival in the experiment: A group of 1 in the process of rearing the right leg scratches,the length of 5cm or so,(estimated to be rabbit cage scratch)infection,to be anti-infective treatment,but the rabbit still died in 4 days,Other New Zealand rabbits did not develop complications such as postoperative infection.2.Compared with blank group A,LVEDD and LVESD were significantly increased and LVEF decreased significantly,P <0.05.3.LVEDD and LVESD were gradually decreased and LVEF recovered gradually at 4 weeks and 8 weeks,and recovered completely at the 12 th week.Compared with the blank group A,P> 0.05.After 1 week of stimulation,After 4weeks,the levels of TGF-β1,Smad2 and Smad3 decreased gradually at 8 weeks,Smad7 increased gradually and recovered at the 12 th week,compared with that before stimulation.Conclusion: Short-card ventricular tachycardia can establish the model of tachycardia cardiomyopathy.The myocardial fibrosis involved in TGF-β1 / Smad signaling pathway is related to the occurrence of cardiomyopathy.The activation of TGF-1,Smad2,Smad3 pathway plays an important role in myocardial fibrosis,and Smad7 plays an inhibitory role.
