Objective(1)To identify the Smilax glabra Roxb.produced in China and Vietnam from character,microstructure and chemical components,and then make sure whether the Vietnam's Smilax glabra Roxb.Can be used for clinical.(2)To establish the high performance liquid chromatography(HPLC)fingerprint method and quantitative analysis of multi-components by single marker(QAMS)method of Smilax glabra Roxb.(3)To research the content change of neoastilbin,astilbin,neoisoastilbin,isoastilbin and engeletin in different growing habitats and years.Methods(1)The UPLC-MS method was used to compare the contents of alcohol extracts in domestic and Vietnam's Smilax glabra Roxb..(2)The HPLC-ELSD method was used to compare the carbohydrates components of domestic and Vietnam's Smilax glabra Roxb..(3)The method of HPLC was used to established fingerprint of Smilax glabra Roxb.and validate the methodology.(4)The method of HPLC was used to establish the method of QAMS and validate the methodology.(5)The morphological identification and microscopic identification method was adopted to compare the structure difference of domestic and Vietnam's Smilax glabra Roxb..Results(1)19 compounds were detected from the alcohol extracts of Smilax glabra Roxb.,and the structure of 18 compounds were elucidated by UPLC-MS,these compounds were determined as:succinic acid,hexadecanoic acid,3,4-dihydroxyphenothyl-3-O-?-D-glucopyr anoside,3,5-dimethoxy-4-hydroxy-benzoic acid-O-?-D-glucoside,(-)-catechin?(+)-catechin,5-O-caffeoylshikimic acid,smiglanin,neoastilbin,astilbin,neoisoastilbin,isoastilbin,engeletin,quercetin,2,4,6-trihydroxyacetophenone-2,4-O-?-D-glucopyranoside?isoengelitin,(+)-Syringaresinol-4-O-?-D-glucopyranose(1?)?-D-glucopyranoside and naringenin.(2)There is no difference in forms of polysaccharides in the domestic and Vietnam's Smilax glabra Roxb.,but in contents,Vietnam's are lower.(3)To establish the high performance liquid chromatography(HPLC)fingerprint method,six common peaks were detected,their similarities were all greater than 0.90,then determined the Vietnam's Smilax glabra Roxb.based on Chinese Smilax glabra Roxb.fingerprints,10 batches of Vietnamese Smilax glabra Roxb.samples similarities were greater than 0.90,there are no significant differences in HPLC fingerprints between domestic and Vietnam's Smilax glabra Roxb..(4)To establish the QAMS method of Smilax glabra Roxb.,it allows simultaneous determination six compounds(neoastilbin,astilbin,neoisoastilbin,isoastilbin and engeletin)and showed good linearities in the range of 0.0362-5.4341 ?g(r2=1.0000),0.0362-5.4341 ?g(r2=1.0000),0.0425-6.3681 ?g(r2=1.0000),0.0372-5.5811 ?g(r2=1.0000)and 0.0068-1.0166 ?g(r2=1.0000),the average recoveries of neoastilbin,astilbin,neoisoastilbin,isoastilbin and engeletin are 93.20%(RSD1.3%),95.39%(2.2%),93.80%(2.7%),96.37%(2.3%)and 99.05%(1.9%).The method shows good repeatability,stability and accuracy.(5)The results showed that there was no obvious contents difference in different growing habtitats of China and have remarkable contents difference between domestic and Vietnam's Smilax glabra Roxb.,the The largest difference chemical component is astilbin.(6)The content of each component in the different growth years was different,the astilbin decreased with the increase of growth years,but the neoastilbin,neoisoastilbin and the isoastilbin have the reverse trend,the contents of five components are varied from plant to plant.(7)There is no significant difference in character and microstructure between domestic and Vietnam's Smilax glabra Roxb..Conclusion(1)The Vietnam's and domestic Smilax glabra Roxb.have no obvious difference except the size,the sections colour.,and the contents,Vietnam's Smilax glabra Roxb.is generally bigger,deeper,have higher contents of flavonols and monosaccharides and have lower contents of Polysaccharides.(2)We have identify 18 chemical compositions and a new element according to the LTQ-Orbitrap-MS,the method of UPLC-MS can be used for the composition analysis and authenticity identification of Smilax glabra Roxb..(3)We determined 55 samples by the method of QAMS and compared with the method of external standard method,the average deflections are less than 1.0%,its indicating that the method have highly accurate and can be used for the quality control of Smilax glabra Roxb..(4)The results showed that there were significant differences in the content of five indicators between different plants and different growth years,the astilbin decreased with the increase of growth years,the neoastilbin,but the neoisoastilbin and the isoastilbin have the reverse trend.
