Objective:Adhesion formation is a normal complication of tendon repair,and normal gliding function can be impaired by excessive adhesion.Tendon adhesion is mainly due to involvement of oxidative stress,while we find that hydrogen-rich water can inhibit oxidative stress.So our study was to investigate if antioxidants hydrogen-rich water can reduce tendon adhesion by antagonizing oxidative stressMethods:(1)First,both Achilles tendons of healthy Sprague Dawley rats were completely ruptured,then sutured by lines.Tendon injury was induced in rats.Sprague Dawley rats were randomly allocated into 2 groups:hydrogen-rich saline group and normal saline group.The rats were induced by intraperitoneal injection.(2)Laboratory tests:1.Serological tests:during postoperative healing,superoxide dismutase(SOD),Malondialdehyde(MDA)and glutathione(GSH)were determined to evaluate oxidative stress and the treatment effects of hydrogen-rich saline.2.Gross morphology and tendon adhesion of each group were determined.3.Tendons were taken for pathology observation by HE staining and Masson staining.4.Biomechanical test:Maximum tensile resistance of tendons were evaluated at 6 weeks after operation.Results:1.Serological tests:The SOD level and GSH level were significantly decreased after operation,while the MDA level was significantly increased.Hydrogen-rich saline led to higher levels of SOD and GSH,while lower level of MDA,as compared with that in the saline solution group.2.Gross finding:Tendon adhesion of each group showed no differences between Hydrogen-rich saline group and normal saline group after 1 week.While after 3 weeks and 6 weeks after operation,adhesion of HS group was significantly better than NS group.3.Tissue morphology:We could find difference degrees of hyperemia edema and inflammatory at 3 weeks after operation.Hydrogen-rich group showed lower level of inflammation.Inflammation was gradually subsided,fibroblast proliferated and disorganized collagen fibers can be seen in fracture.Compared to NS group,collagen fibers of HS group showed better arranged.4.Compared to normal saline group,Hydrogen-rich saline group showed better on maximum tensile resistance.Results:1.Tendon injuries can led to oxidative stress,while Hydrogen-rich saline can reduce it.2.Hydrogen-rich saline can reduce the extent of adhesion of healing tendons,probably by redox modulation,in a rat model.
