Objective:This study attempt to detect prostaglandin E2(PGE2),apolipoprotein al(APOA1),Kininogen-1(K1),Retinol-binding protein(RBP4)in vitreous of rhegmatogenous retinal detachment associated with choroidal detachment(RRDCD),rhegmatogenous retinal detachment(RRD)and idiopathic epimacular membrane(IEM),evaluating the predictive potential of PGE2,APOA1,K1,RBP4 for RRDCD.Methods:We used Enzyme linked immunosorbent assay(ELISA)to analysis the level of prostaglandin E2 in 56 samples from patients(26 RRDCD and 30 RRD)undergoing pars plana vitrectomy.We used ELISA to obtain the levels ofAPOAl,Kl,RBP4 from the vitreous samples of 76 primary patients included 23 RRDCD patients,28 RRD patients,24 IEM patients undergoing pars plana vitrectomy.The differences of molecular biomarkers concentration between groups were analyzed using the Mann-Whitney U-test/Kruskal-wallis for nonparametric values or independent samples T test/one-way ANOVA analysis for measurement data.The relationship between mediators and the grades of PVR and quadrants of retinal detachment were analyzed using nonparametric Spearman rank correlation analysis.Results:1.The level of PGE2 in vitreous of RRDCD was less than RRD,and the difference was statistically significant(P=0.000).The level of PGE2 in vitreous of RRDCD were no significant difference between defferent grades of PVR(A,B,C,D)(P=0.409).The level of PGE2,PGF2a in vitreous of RRDCD were no significant difference between mild PVR(A,B)and heavy PVR(C,D)(P=0.433).PGE2 increased with the number of retinal quadrants detached in RRDCD but nonsignificantly.2.The ELISA outcomes documented that the concentration of APOA1?RBP4 in vitreous were significantly higher in RRDCD than RRD and IEM.The concentration of K1 were not significantly different between three groups.The concentration of APOA1?RBP4 in vitreous were higher in severe PVR patients than those in mild PVR with no statistical significant difference.The concentration of K1 in vitreous were higher significantly in mild PVR patients than those in severe PVR.In RRDCD group,APOA1 is positively correlated with RBP4 and negatively correlated with K1 with statistical significant difference.RBP4 is negatively correlated with KI with no statistical significant difference.In RRD group,RBP4 is positively correlated with APOAland negatively correlated with Klwith statistical significant difference.APOA1 is negatively correlated with K1 with no statistical significant difference.Nonparametric Spearman rank correlation analysis revealed that APOA 1 and RBP4 increased significantly with the number of retinal quadrants detached in RRDCD and RRD groups.K1 decreased with the number of retinal quadrants detached in RRDCD groups with no statistical significance difference,while Kl decreased with the number of retinal quadrants detached in RRD groups with statistical significance.Conclusion:1.The level of PGE2 in vitreous of RRDCD was less than RRD and consistent with earlier results.It might might indicate a greater consumption of PGE2 that would be stimulated by inflammation or a loss of limitation of inflammatory responses.Besides,the level of PGE2 did not correlate with PVR grades and the number of retinal quadrants detached.2.APOA1 and RBP4 were significantly upregulated in the vitreous of RRDCD eyes compared with RRD group and IEM group.APOA1 and RBP4 were increased significantly with the number of retinal quadrants detached in RRDCD group.APOA1 and RBP4 has a tendency to increased with the severity of PVR(but nonsignificantly).The findings of this study may display the potential of APOA1 and RBP4 as specific biomarkers of RRDCD.However,the concentration of K1 displayed a weaken potential as specific biomarkers of RRDCD.The findings of this study may contribute to increased understanding regarding the role of PGE2,APOA1,RBP4 and K1 in RRDCD.
