Postoperative ileus(POI)is a self-limiting defense response of the body itself to the surgical trauma,which is an unavoidable complication in surgical operation.The primary features of POI include nausea,vomiting,inability to tolerate an oral diet,abdominal distension and delayed exhaust and defecation.Postoperative ileus has be show slow patient recovery,prolonged hospital stay,and also can lead to a variety of postoperative complications increased incidence.Postoperative intestinal obstruction not only increased the pain on patients,but also increased the psychological burden of patients,increasing the difficulty of postoperative recovery.At the same time,POI caused by the over cost of treatment and medical resources wasted,it’s a huge medical expenses.In recent years,the study of POI of the heat gradually increased,study shows that postoperative intestinal inflammatory response is the key to POI.It has become an important reacharch method to use of gene knockout technology of building a comparative study for the pathogenesis of POI research and the mechanism of the body’s gastrointestinal protection.Steroid receptor coactivator 3 is a member of the coregulator SRC family,which interacts with a variety of transcription factors and their corresponding nuclear receptors,to enhance their transcriptional activity.In recent years,Yu C and its team have found that SRC-3 knockout can lead to excessive inflammatory response.In the previous experiment,we successfully constructed the model of POI disease in mice and found that after the intestinal obstruction,expression of SRC-3 protein in the intestinal muscle of mice was increased.Comparative study between the POI model mice of SRC-3 knockout mice and wild type mice show that the expression in mRNA and protein of inflammatory cytokines and chemokines in the intestinal mucosa of SRC-3 knockout mice is higher is than that of Wild type mice.Recently,we found that the intestinal inflammatory cell infiltration of SRC-3 gene knockout mice after surgery model mice after was significantly more than that in wild-type mice.These results suggest that SRC-3 may play an immunomodulatory role in the over-inflammatory response in ileus,thereby protecting the intestinal tract in POI.
