| Objective:1.LAMP3,CD1a,CD83 is the detection more reliable immune markers of dendritic cells(DCs),LAMP3,CD1a and CD83 expression level in tumor tissue samples from patients were detected in simple thymoma and myasthenia gravis associated with thymoma,explore dendritic cells related role for expression in myasthenia gravis associated with thymoma patients in immune pathogenesis process;and further explore the expression of dendritic cells between the patients with Osserman type,Masaoka staging and related to the association clinical pathological factors.2.To compare the differences of the expression of CD1a and CD83 in the tumor tissues of patients with thymoma and myasthenia gravis,and to explore the correlation between immature dendritic cells and mature dendritic cells.3.The expression level of LAMP3,CD1a and CD83 in tumor patients with simple thymoma and thymoma associated with myasthenia gravis were used to detect,through the analysis of test results,to further broaden the understanding of immunity on the pathogenesis of MG,which is beneficial to the pathogenesis of more in-depth research,so as to provide more useful ideas and new treatment for clinical practice.Method:37 cases of thymoma associated with MG and 13 cases of thymoma were selected.All patients were not treated with radiotherapy,glucocorticoid and immunosuppressive agents.Methods immunohistochemical staining(SP method)to detect dendritic cell immune markers LAMP3,CD1a,CD83 in tissue samples of pure thymomas and thymomas with two groups of patients with MG expression,and through the application of statistical analysis software package SPSS20.0 t test andχ~2 test statistical difference between the correlation results in,Spearman rank correlation analysis method to analysis the correlation between LAMP3 and CD1a,LAMP3 and CD83,CD1a and CD83 of the two inspection standards set for P≤0.05.Results:1.The positive expression of LAMP3 protein in thymoma tissue of MG patients was73%(27/37),the positive expression in tissues of patients with simple thymoma was38.5%(5/13),the difference was statistically significant(P<0.05).The positive expression of LAMP3 was closely related to the pathological type of WHO,and B1and B2 were the most significant.And its expression was significantly associated with thymoma Masaoka,with the increase of staging of thymoma showed Masaoka positive expression rate decreased,stage IV expression is lowest,the difference was statistically significant(P<0.05).2.The positive expression of CD1a protein in thymoma tissue of MG patients was86.5%(32/37),the positive expression in tissues of patients with simple thymoma was53.8%(7/13);the difference was statistically significant(P<0.05).The positive expression of CD1a protein was closely related to the pathological type of WHO,and B1 and B2 were the most significant.And its expression was significantly associated with thymoma Masaoka,with the increase of staging of thymoma showed Masaoka positive expression rate decreased,stage IV expression is lowest,the difference was statistically significant(P<0.05).3.The positive expression of CD83 protein in thymoma tissue of MG patients was64.9%(24/37),the positive expression in tissues of patients with simple thymoma was38.5%(5/13),the difference was statistically significant(P<0.05).The positive expression of CD1a protein was closely related to the pathological type and Masaoka stage of thymoma,and the positive expression rate of WHO protein was decreased with the increase of Masaoka stage.4.In the WHO pathological typing of thymoma tissues of each group,through the comparison of CD1a and CD83 in thymoma and simple thymoma patients with MG were found the CD1a expression level of staining scores were better than CD83protein positive expression rate and the average cell expression situation.The expression levels of LAMP3 and CD1a,LAMP3 and CD83 in thymoma with MG were parallel,and there was a positive correlation.5.There is some relationship with CD1a,LAMP3 and CD83 in thymoma with expression of Osserman and MG in tissue typing,the expression is better than that of type III and type IV three combined type,I Osserman MG in type IIA,IIb type in thymoma,type IV is the lowest;but the three indexs and the difference between the groups of Osserman type was not statistically significant(P<0.05).Conclution:1.By comparing the simple thymoma group,combined with immunotherapy of dendritic cell marker MG test group tissue LAMP3,CD83 and CD1a expression in thymoma and thymic tumor Masaoka staging and WHO pathological type is closely related to that in thymoma with expression of dendritic cells in MG tissue was higher than that in thymoma,suggesting increased dendritic cells the abnormal expression may be one of important factors in the occurrence and development of the process involved in the pathogenesis of MG.2.In the cases of thymoma with MG tissue and simple thymoma group,CD1a protein positive expression rate and the average cell staining scores were higher than those of CD83 protein,suggesting that mature dendritic cells and immature dendritic cells are involved in the pathogenesis of MG,and the expression of immature dendritic cells than mature dendritic cells expression.Immature dendritic cells in the pathogenesis of MG plays a more active role in leading.3.There is some relationship with CD1a,LAMP3 and CD83 in thymoma with expression of Osserman and MG in tissue typing,reduce with Osserman type level increasing trend,but Osserman difference of the three factors between groups was not significant statistically,there may be a certain relationship with the quantity of samples for research small and subjectivity Osserman typing,further research is necessary. |
