Objectives: To establish a mass spectrometry for comparing the differences proteins in Uyghur type 2 daibetes mellitus(T2DM)and normal people;to identify differentially expressed proteins related to Uyghur T2 DM,to provide the basis for revealing the pathogenesis of Uyghur T2 DM,and to predict the ocurrance of diabetes complications.Methods: Forty six patients with T2 DM and normal control human serum were collected.Two groups of serum samples were combined into four groups,a total of eight groups of sample pool.iTRAQ(isobaric tag for relative and absolute quantitation)combined with LC-MS/MS(nano liquid chromatography tandem mass spectrometry)was used to isolate differential expression proteins of Uyghur T2 DM and normal control group.Search and identification were performed by using the Mascot and Uniprot-human database,then the relevant proteins that meet the requirements were selected,and bioinformatics analysis were carry out.Results: Compared with the normal control group,the protein expression of serum in Uyghur T2 DM patients was different.According to the screening conditions,59 differentially expressed proteins(fold change Log2?|0.6|)were screened for the T2DM-related protein,which was 45 kinds of up-regulated proteins and 14down-regulated proteins,the difference was statistically significant(P <0.01).GO analysis revealed that these proteins were mainly located in the cell part(19.0%),organelles(15.5%)and extracellular(13.1%);Biological processes involving cellular processes,metabolic processes,cellular components,biosynthesis processes,immunization system,biological regulation and so on;Molecular functions include protein binding,catalytic activity,structural molecular activity,antioxidant activity,transport protein activity,receptor activity.PHNTHER Pathway analysis showed that the differential protein was mainly involved in ? cell activation,T cell activation,blood coagulation,DNA replication,heterotrimeric G protein signal transduction pathway,integrin signaling pathway,and some differential proteins involved in glycolysis,Epidermal growth factor receptor signaling pathway,vitamin D metabolism pathways and other pathways.The PPI interaction network of differential proteins was constructed.The proteins at the nodes were SERPINAI,CP,KNG1,TF,ALB,GAPDH,LTF,ACTB,YWHAZ,GSN,APOA4 and so on.The occurrence of T2 DM might be related to these proteins.Conclusion: There are significant differences in serum protein between Uyghur T2 DM patients and normal subjects.A variety of serum biomarker candidate proteins associated with Uyghur T2 DM patients were identified and established a foundation for further validation of biomarkers.
