Objective: To investigate the relationship between Single Nucleotide receptor(GHR)gene Growth hormone Polymorphism(SNP)and genetic susceptibility of hepatocellular carcinoma(HCC)in liver cancer family pedigress of Fusui County of Guangxi province.Methods: In this subject,twenty-five Guangxi high-incidence families of hepatocellular carcinoma(including 25 patients withHCC and 81 members of the immediate family of HCC)and 17 control families(80 subjects in the control group)were used as subjects.The GHR gene was detected in two groups using time-of-flight mass spectrometry.Genotypic and allelic frequencies of rs6451620 loci were used,and a non-conditional Logistic regression model was used to analyze the relationship between this locus polymorphism and genetic susceptibility to hepatocellular carcinoma.Results: 1.There were two alleles of A and G,and three genotypes of AA,GA and GG in rs6451620 locus of GHR gene.2.The genotype frequency of GHR gene rs6451620 locus was in accordance with the genetic balance,and itwas representative of the Zhuang Population in Guangxi.3.The frequencies of GHR genotype rs6451620 A and G allele frequencies were 36% and 64% in liver cancer group,and 39.5% and 60.5% respectively in liver cancer family non-liver cancer group.They were distributed in healthy control pedigrees.The frequency is 29.4% and 70.6%.The GHR gene rs6451620 locus was 0.806 times(95%CI 0.27-2.41)in individuals carrying the A allele of the liver cancer family non-hepatoma group,and the risk was not statistically different from the individuals carrying the G allele;Individuals carrying the A allele in the healthy control pedigree had a 1.488 times higher risk of liver cancer than those with the G allele(95% CI: 0.73-304,P=0.274),which means the difference is not statistically significant.The HBV-negative GA+AA genotype of GHR gene rs6451620 was 4.671 times that of GG genotype(P>0.05,the difference was not statistically significant).The GA+AA genotype carrying HBV-positive individuals was 2.155 times higher than that of GG genotypes,with no significant difference(P>0.05).Conclusion: Two genotypes G and A and three genotypes AA,GA and GG existed in the rs6451620 locus of GHR gene.There were no significant correlations between GHR gene rs6451620 locus and the two genotypes of GHR gene and the genetic susceptibility of liver cancer in Guangxi Fusui.In the sample population,the risk of genotype AA carrying the rs6451620 genotype of the GHR gene was not significantly different from the genotype GG carrying the risk of HCC.
