Periostin can remodel the metastatic niche,stem cell niche,perivascular niche and pre-metastatic niche to facilitate the progression of solid tumor,while no one has reported its function in hematological malignancies.Here we first investigated its role in the progression of B-ALL(B-cell acute lymphoblastic leukemia).We found the expresstion of periostin was drastically elevated in the bone marrow of human and mouse B-ALL conditions.Knocking out periostin ameliorated the leukemia burden by inhibiting the proliferation and metastasis of B-ALL cells.In vitro experiments showed that the recombinant periostin treatment could promote the proliferation and colony formation ability of B-ALL cells.Using in vivo sorting experiments and in vitro co-culture experiments,we also demonstrated that the elevated periostin in B-ALL conditions was secreted by BM-MSCs(Bone marrow-derived mesenchymal stomal cells).Further mechanism study showed periostin could activate ERK pathway to promote the proliferaton of B-ALL cells.Our study first indicated periostin as a new factor in bone marrow to promote the progression of B-ALL.
