| Objective:1.Model the rat model of liver stagnation and kidney deficiency syndrome caused by chronic unpredictable mild stress;2.Based on β-endorphin to explore the mechanism of chronic unpredictable mild stress induce liver stagnation and kidney deficiency syndrome in rats.Method:By chronic unpredictable mild stress,model the rat model of liver stagnation and kidney deficiency syndrome.according to the method of random number table,20 Wistar male rats were randomly divided into normal group and model group,and 10 in each group.through the binding,the ice water swimming and other methods,observe the rat’s general changes,and weigh the rat’s body weight,wet weight of liver,spleen,kidney and adipose;the behavioral changes of rats were analyzed by the sucrose water consumption test,open field test and the water maze experiment;the indicators of liver stagnation:the serum DA,CRH and CORT content of HPA axis were detected by Elisa,and the indicators of kidney deficiency were detected:serum 17-OHCS,GH,ALD,EPO and T content;the serum GnRH,FSH,LH content of HPG axis and hypothalamus β-EP content were detected,and the gene expression level of Oprm in the hypothalamus was detected by real-time PCR.Results:Compared with normal group,model group rats showed yellow hair and shrill voice,depressed,tired,lazy and unresponsive.rat’s body weight,wet weight of spleen,liver,kidney and adipose significantly decreased(P<0.01);the sucrose water consumption reduced(P<0.01).the total distance,the average velocity and central distance of OFT decreased significantly(P<0.01).the average time and average distance increased from four different entrances to platform in the water maze experiment(P<0.01);the serum DA content decreased(P<0.05),and the content of CRH and CORT significantly increased(P<0.01),and the content of serum GH,ALD,T,EPO and 17-OHCS decreased(P<0.01);serum GnRH,FSH and LH content significantly reduced(P<0.01),and the content of hypothalamus β-EP significantly increased(P<0.01),and the gene expression level of Oprm in the hypothalamus increased(P<0.05).Conclusion:1.Chronic unpredictable mild stress can model the rat model of liver stagnation and kidney deficiency syndrome.2.The pathogenesis of liver stagnation and kidney deficiency syndrome induced by chronic unpredictable mild stress may be closely related to the involvement of β-EP in the regulation of HPA axis and HPG axis. |
