Objective:Dendrobium officinale,a perennial epiphytic herb from Dendrobium,is a traditionally precious Chinese herbal medicine,and is also a commonly used fresh medicine."when traditional Chinese medicine uses Dendrobium officinale,it pays attention to fresh products for superiority".Compared with dry products,fresh products have the characteristics of strong moisturizing dryness properties.However,due to the high cost of storage and transportation of fresh Dendrobium officinale and the limitation of processing technology,fresh Dendrobium officinale is difficult to keep fresh,which limits its clinical application and is mainly replaced by dry products in clinical.In this paper,Dendrobium officinale as the research object firstly investigate the influence of different drying methods on total polysaccharide,mannose content and antioxidant activity to screened out the most suitable drying method.On this basis,through optimizing the molding process parameters of granules,the fresh Dendrobium officinale granules with convenience for storage,transportation and use was prepared.To further study the quality standards of fresh Dendrobium officinale granules,a method for its quality control was established.Methods:The influence of four drying methods,such as drying,infrared,microwave and vacuum freezing,on polysaccharide content and antioxidant activity of Dendrobium officinale were investigated.The establishment of phenol-sulfuric acid method for the determinationoftotalpolysaccharidecontent,highperformanceliquid chromatography?HPLC?for mannose content and DPPH·,ABTS·+,·OH radical scavenging test,the optimal drying method of Dendrobium officinale was obtained;With the qualified particle yield,angle of repose,and moisture absorption rate as evaluation indexes,the process parameters such as dosage of medical dextrin,ethanol concentration,drying temperature,and drying time were optimized through the single-factor test and BBD method to confirm the optimal formulation ratio of fresh Dendrobium officinale granules,optimize its preparation process and research the preparation of fresh Dendrobium officinale granules;The quality control methods of fresh Dendrobium officinale granules were established by selecting the appearance,granularity,solubility,moisture content,loading amount difference,identification and content determination?total flavonoid content?to provide a reasonable,standardized and scientific quality control system for fresh Dendrobium officinale granules.Results:1.Investigation and comparison of the optimal drying methods for Dendrobium officinaleThe influence of different drying methods on polysaccharide and antioxidative activity of Dendrobium officinale were investigated by taking total polysaccharides,mannose content and antioxidant activity as indexes.Comprehensive evaluation of the quality of Dendrobium officinale treated by different drying methods was in sequence:vacuum freeze drying>microwave drying>oven drying>infrared drying,Among them,the content of total polysaccharides and mannose in vacuum freeze-dried Dendrobium officinale was the highest,which was 431.6?20.57 and 383.2?35.14mg·g-1,respectively,and the scavenging effect of DPPH·?ABTS·+?and·OH was the strongest.The results showed that the vacuum freeze drying was significantly better than the other three kinds of drying methods?p<0.05?,more beneficial to retain the antioxidant activity of Dendrobium officinale.2.Screening and optimizing the optimal preparation process of fresh Dendrobium officinale granulesTaking the dosage of medical dextrin,ethanol concentration,drying temperature,and drying time as inspection factors,the qualified particle yield,angle of repose,and moisture absorption rate as the evaluation indexes,and referring to the Box-Benhnken Design?BBD?Center-combined experimental design principle,the formulation ratio was determined by response surface analysis.The optimal preparation process for screening fresh Dendrobium officinale granules is as follows:dosage of medical dextrin is 70%,ethanol concentration is 50%,drying temperature is 40?and drying time is 3h.Under this prepared process,Critical relative humidity of fresh Dendrobium officinale granules is 50%,angle of repose is 25.6 degrees and bulk density is0.55g/ml,indicating that the obtained granules have good quality and formability and accord with the requirements of pharmacopoeia.3.Study on quality standards of fresh Dendrobium officinale granulesSelect quality control indicators such as appearance,granularity,solubility,moisture content,loading amount difference,identification,content determination?total flavonoid content?to establish quality standards for fresh Dendrobium officinale granules.The results showed that TLC identification and content determination of fresh Dendrobium officinale granules were simple,specific,and the quality standards are stable and controllable,which can effectively control the quality of fresh Dendrobium officinale granules.Conclusion:Compared with other drying methods,vacuum freeze-dried Dendrobium officinale has the strongest antioxidant effect,which can retain the total polysaccharides and mannose of Dendrobium officinale to the utmost extent;The optimal preparation process of fresh Dendrobium officinale granules was obtained by response surface analysis.According to the characteristics of fresh Dendrobium officinale granules,the quality standards of its were preliminarily established.The study confirmed that the parameters of the established quality standards were stable and controllable,and can effectively control the quality of fresh Dendrobium officinale granules.These studies have laid a solid foundation for the development of fresh Dendrobium officinale granules.
