Objective: To study the expression of high mobility group protein B1(HMGB1),proliferating cell nuclear antigen(PCNA)and matrix metalloproteinase 9(MMP-9)observed in esophageal squamous cell carcinoma tissues,para-carcinoma tissues and normal esophageal tissues,and analyze their relationship with clinical pathological features in esophageal squamous cell carcinoma.The concentrations of HMGB1,PCNA and MMP-9 in the serum of preoperative patients with esophageal squamous cell carcinoma and benign esophageal diseases were observed,in order to find new tumor marker for diagnosis of esophageal squamous cell carcinoma.Methods: Immunohistochemistry and RT-PCR were used to detect the expression of HMGB1,PCNA and MMP-9 in 89 cases of esophageal squamous cell carcinoma tissues,30 cases of para-carcinoma tissues and 30 cases of normal esophageal tissues,and the clinical pathological data of esophageal squamous cell carcinoma were analyzed.Enzyme linked immunosorbent assay(ELISA)was used to detect the concentrations of HMGB1,PCNA and MMP-9 in peripheral blood serum of 30 cases of esophageal squamous cell carcinoma and 30 cases of benign esophageal lesions.Results: The expression of HMGB1,PCNA and MMP-9 in esophageal squamous cell carcinoma tissues were significantly higher than those in adjacent tissues and normal esophageal tissues(P<0.05).There was no significant difference in the expression rate between adjacent tissues and normal esophageal tissues(P > 0.05).The expression of HMGB1 and MMP-9 in esophageal squamous cell carcinoma was not related of sex,age and pathological differentiation degree(P > 0.05),which were related to the depth of invasion,lymph node metastasis and TNM stage(P<0.05).The expression of PCNA in esophageal squamous cell carcinoma was not related to the sex and age of patients(P > 0.05),but was correlated with pathological differentiation degree,the depth of invasion,lymph node metastasis and TNM stage(P<0.05).The expression of PCNA in the tissues of squamous cell carcinoma of the esophagus was positively correlated with the expression of MMP-9(r=0.8751,P<0.05).The expression of HMGB1 in the tissues of squamous cell carcinoma of the esophagus was positively correlated with the expression of PCNA(r=0.9358,P<0.05).The expression of MMP-9 in the tissues of squamous cell carcinoma of the esophagus was positively correlated with the expression of HMGB1(R =0.7631,P<0.05).The serum concentrations of HMGB1 and MMP-9 in esophageal squamous cell carcinoma were significantly higher than those in patients with benign esophageal lesions(P<0.05),While here was no significant difference in the expression of PCNA in serum.A positive correlation was discovered between the expression of HMGB1 and MMP-9 in the carcinoma tissues and their concentrations in serum(P<0.05).Conclusion: The clinical detection of HMGB1,PCNA and MMP-9 in esophageal squamous cell carcinoma might be helpful to diagnosis and prognosis of esophageal squamous cell carcinoma.HMGB1 and MMP-9 concentrations in peripheral blood might be helpful to the early diagnosis of esophageal squamous cell carcinoma,but PCNA is not be recommended for peripheral blood concentration.
