| This paper selected two marine-derived fungi,one is Aspergillus sp.XT-20-8 from the deep eastern Pacific Ocean,and another is Trichoderma atroviride H548 from mangrove,to study the active ingredients.For T.atroviride H548,through metabolomics and HPLC analysis,we investigated the effect of seven epigenetic regulators on metabolomes,5-azacytidine was selected as an epigenetic intervention molecule to add to the medium for fermentation,to look for the differential secondary metabolites with wild type T.atroviride H548.There,15 compounds have been isolated,3 compounds are novel a-cyclopentenone acrylic acid derivatives,4 compounds are known cyclopentenone acrylic acid derivatives,3 compounds are terpenes and 1 compound is chromogen dimer,3 differential metabolites found are 3-(3-oxocyclopent-l-en-l-yl)propanoic acid,(E)-3-(3-oxocyclopent-1-en-1-yl)acrylic acid and methyl-3-(3-oxocyclopent-1-en-1-yl)propanoate.For the separation of Aspergillus sp.XT-20-8 metabolites,firstly,we used the conventional separation method,silica gel column-ODS column-preparative liquid phase chromatography separation,15 compounds have been isolated.4 compounds are identified as the cyclic peptide,3 compounds are sesquiterpenoids derivatives,and 2 compounds are nucleoside derivatives,4 compounds are resorcinol dimers.Then,we constructed a new separation system,the new normal phase liquid chromatography × reversed phase liquid chromatography separation,we obtained 12 compounds corresponding with conventional separation from Aspergillus sp.XT-20-8 extract.These compounds were tested cytotoxic activity,and found that compound 37 exhibit moderate activity on cervical cancer cell Hela-S3,esophageal cancer cells ECA-109 and hepatoma cells BEL7402.In this paper,we separated 26 compounds from T.atroviride H548 and Aspergillus sp.XT-20-8,3 novel compounds among them.And we constructed a newly preparative NPLC×RPLC system,which can be applied to the separation of complicated natural products. |
