Background and objectives:Breast carcinoma is the most common malignant tumors in women.Although in recent years,various regions of China actively carry out breast cancer screening work,and actively carry out various preventive measures,the incidence of breast carcinoma is increasing year by year,and it tends to be younger.As related to tumor molecular biology research,and biological applications of molecular techniques and the improvement of clinical diagnosis and treatment,diagonosis and treatment of breast carcinoma had great progress,significantly improved the survival rates of patients with breast carcinoma,but the heterogeneity of breast carcinoma has caused a lot of problems remain to be solved.In recent years,related studies have shown that breast carcinoma stem cells play an important role in the development of breast carcinoma.The experiment of which have been proven for breast carcinoma stem cell markers ofacetaldehyde dehydrogenase 1(acetaldehyde dehydrogenase 1,ALDH1)and stromal cell derived factor receptor 4(CXC chemokin receptor 4,CXCR4)expression in breast carcinoma research,analysis ALDH1 and CXCR4 expression weiht different molecular subtypes of breast carcinoma the relationship between the indirect to explore the role of cancer stem cells in breast carcinoma.Meanwhile,studies have pointed that micro ribonucleic acid(microRNA,miRNA)involved in the regulating of cell proliferation and apoptosis,and plays an important role in the occurrence and development of tumors.It is similar to the functions of tumor suppressor gene and oncogenes.Among them,them,the expression of micro ribonucleic acid 21(miRNA-21)in breast carcinoma may be related to the regulation of stem cell proliferation and apoptosis.At the same time,the expression of miRNA-21 in breast carcinoma tissue in this experiment,analysis of the expression of mi RNA-21 in the relationship between different molecular subtypes of breast carcinoma,and correlation with tumor stem cells ALDH1 and CXCR4 expression in breast carcinoma tissues,,on this basis to further explore the regulation relationship between miRNA-21 and tumor stem cell.Materials and methods:Collection of Basic Medical College of Dalian Medical University from 2011 to 2013,24 cases of invasive ductal carcinoma,and China Medical University Affiliated Hospital from 2016 to 2018 Dalian Shengjing invasive ductal carcinoma in 22 cases,a total of 46 cases of breast infiltrating ductal carcinoma tissues.According to the expression of ER,PR,HER-2 and Ki-67 all breast carcinoma patients were divided into Luminal A,Luminal B,HER-2 over expression of type Triple Negative and 4 subtypes,including 19 cases of type Luminal A,9 cases of type Luminal B,HER-2 over expression of type 7 cases,11 cases of type Triple Negative.Immunohistochemical staining(SP method)to detect the expression of ALDH1 and CXCR4 in breast carcinoma tissues and to detect the expression of mi RNA-21 by in situ hybridization in breast carcinoma and the expression of miRNA-21 and cancer stem cells after the combination of ALDH1 and CXCR4 in different breast carcinoma molecular subtypes of tumor,analysis the relationship between the molecular subtypes of stem cell and breast carcinoma,and cancer stem cells miRNA-21 and ALDH1 and CXCR4 in breast: correlation between the expression of different molecular subtypes.The experimental data were analyzed by SPSS 19 software,using the statistical software of rank and inspection,different molecular subtypes of breast carcinoma of miRNA-21,a positive ALDH1 and CXCR4 expression in comparison,comparing two with variance analysis method.P<0.05 indicates a statistical significance.Results:1.The expression levels of ALDH1 and CXCR4 were different between different molecular subtypes of breast carcinoma.Among them,over expression of HER-2 ALDH1 and CXCR4 type and Triple Negative type medium showed a strong positive expression level was higher than that of Luminal A and Luminal B(P<0.05),and over expression of HER-2 and Triple Negative expression,there was no significant difference(P>0.05).2.There is a difference in the expression level of miRNA-21 between different subtypes of breast carcinoma.Among them,over expression of HER-2 type and Triple Negative type miRNA-21 medium positive Xeon expression levels higher than that of Luminal A type and Luminal type B(P<0.05),and between the expression of HER-2 and Triple Negative showed no significant difference(P>0.05).3.The positive expression of miRNA-21 high degree of over expression of HER-2 and Triple Negative,the positive expression of ALDH1 and CXCR4 levels were significantly higher,and significantly higher than that of Luminal A and Luminal B,the high expression of miRNA-21 molecular subtypes,ALDH1 and CXCR4 also showed high expression of miRNA-21,low expression of molecular subtypes ALDH1 and CXCR4 also showed low expression.Conclusions:1.The expression level of tumor stem cell ALDH1 and CXCR4 in different molecular subtypes of breast carcinoma tissues is different,and participates in the development and development of different molecular subtypes of breast carcinoma.2.The expression level of miRNA-21 in breast carcinoma tissues with different molecular subtypes is different,which is involved in the occurrence and development of different subtypes of breast carcinoma.3.miRNA-21 plays a certain regulatory role in ALDH1 and CXCR4 of tumor stem cells,and may promote the proliferation and differentiation of breast carcinoma stem cells.
