Objective: To find the optimal combination of flavonoids and volatile oils in seeds of NGS to anti-COPD,to explore their effects on the mechanism of COPD.Methods: 1)To observe the acute toxicity of NFVC by fixed dose method;2)Injection of carrageenan in mouse toes,to observe the effect of NFVC on the swelling of the toes in mice;Injection of Freund's adjuvant in rat toes,to observe the effect of NFVC on the swelling of the toes in rat;By using ELISA to detect VONG to LPS stimulation of monocyte / macrophage RAW264.7 mice secrete IL-8,IL-6,the influence of by Griess Reagent method to detect RAW264.7 cells that nitric oxide in the supernatant fluid(NO)concentration.Level of animal experiments;Through the guinea pig isolated smooth muscle experiment,to observe the effects of the NFVC causesd by Ach and His in ileum smooth muscle tension of guinea pig.3)by intratracheal instillation of LPS plus inhalation of cigarette for making rat COPD model,to observe the general situation of rats,tissue pathologic changes,inflammatory cells in BALF and blood were classificatied,levels of IL-8?IFN-??IL-4?IL-17?IL-10?TGF-?in BALF and blood were measured;levels of ?1-antitrypsin?fibronectin in blood were measured,tissue pathologic changes,T lymphocytes cells in blood were classificatied by Flow cytometry.Results: 1)NFVC has no toxicity;2)Compared with group of flavonoids and volatile oil: The concentration 200: 40mg/kg of NFVC decreased the swelling of the foot in mice significantly(P < 0.01);The concentration1 40: 10mg/kg?140: 20mg/kg of NFVC Group had a good therapeutic effect on adjuvant arthritis and had a significant inhibitory effect on immune function(P<0.01);The concentration 80: 12.5?g/ml?80: 25?g/ml of NFVC can inhibit the excessive secretion of RAW264.7 cells,IL-6,IL-8 and NO and a certain dose effect relationship(P<0.01).The concentration 1: 10mg/ml?2: 5mg/m of NFVC could reduce the tension of guinea pig bronchus smooth muscle contraction induced by Ach and His significantly(P<0.01);3)Compared with group of flavonoids and volatile oil: the general condition of rats was improved,the pathologic changes of the lung tissue were reduced in different degree;the volatile oil and flavonoid glycoside group of Leukocytes cells,neutrophils cells,monocytes cells,eosinophils cells,basophils cells,lymphocytes cells percentage in BALF and blood were significantly decreased(P<0.05),levels of IL-8?IFN-??IL-4?IL-17?IL-10?TGF-? in BALF and blood were decreased(P<0.05).Conclusion: Compared with group of flavonoids and volatile oil,NFVC has a good anti-COPD effect.
