Periodontitis is one of the chronic inflammatory diseases.Periodontal pathogens such as Pg with a variety of virulence factors can stimulate the host immune response and cause alveolar bone resorption,and ultimately lead to periodontal tissue damage.It was reported that Pg can escape from the immune of the host,and breed in periodontal tissues,they can secrete some factors which can lead to periodontal inflammation and periodontal tissue destruction.Not only does the harm severely affect chewing,pronunciation,but also affected the living quality of patients.So,it is important to find a sustainable solution to protect the alveolar bone.The recent research find that miRNAs can be the target of periodontitis.Researchers believe that mi RNAs can active the immune cells.mi R-34a is one of these.mi R-34a,residing at 1p36,is a kind of endogenous non-coding RNA with regulatory function.Recently,miR-34a played an important part in inflammatory response and process of bone metabolism.It is reported that miR-34a expression in periodontitis tissue or healthy tissue is different,so we assume that miR-34a can inhibit the alveolar bone absorption.Our experiment aim to find how miR-34a influence the NLRP3、Caspase-1 and IL-1β mRNA expression of BMSCs which is infected by the Pg.Methods:1 Extract the P0 BMSCs from the distal rat femur,and develop them to 3-6 generations.2 Test the transfection efficiency by Flow cytometric method.3 N-Ac-L-Leu-PEI/miR-34a.4 Use Pg to infect the BMSCs after transfect the mir-34a into BMSCs,then detect the mRNA expression change of NLRP3,Caspase-1and IL-1β in qRT-PCR.Results:To compare with the control group,mi R-34a can increase the expression of NLRP3,Caspase-1,IL-1β mRNA.And to compare with the Pg group,the NLRP3,Caspase-1,IL-1β mRNA expression decreased after transfecting miR-34a.Conclusion:miR-34a can trigger the inflammatory response reaction in healthy cells,but miR-34a inhibit the NLRP3,Caspase-1,IL-1β mRNA expression in cells infected by Pg.
