Background and objective The incidence and mortality of lung cancer remain high.The incidence and mortality of lung cancer in Xuanwei,Yunnan are much higher than those in other parts of the country.This is closely related to the local coal-fired pollution,and the disease shows significant family aggregation.The purpose of this study was to explore the potential association between long non-coding RNA(LncRNA)and family lung cancer in Xuanwei district,providing theoretical basis and experimental basis for genetic susceptibility study and early diagnosis and early treatment of lung cancer in Xuanwei district.MethodsIn the previous investigation of family cancer history of lung cancer patients in Xuanwei area,and the Genome Wide Association Studies(GWAS)data analysis of representative lung cancer patients,a high-frequency top ten long non-coding RNA was selected.Primers were designed for high frequency mutation sites.Detection of the expression level of target LncRNA in cancer tissues and adjacent normal tissues of different non-small cell lung cancer patients by qRT-PCR.The expression of target LncRNA was verified in non-small cell lung cancer cell lines A549,NCI-H1299,H1650,HCC-827 and human normal bronchial epithelial cell line BEAS-2B;the key LncRNAs with significant differences in the expression of lung cancer were analyzed.The correlation between clinicopathological features of lung cancer patients provides theoretical and experimental evidence for the further diagnosis and treatment of non-small cell lung cancer.Result1.Xuanwei District Lung Cancer Family top 10 LncRNAs with mutation frequencies:Linc00842,Linc00343,Linc00507,Linc00433,LincC00578,Linc00544,Linc00320,Linc00624,Linc00665,Linc008712.The expression of Linc00665,Linc00624,Linc00320 in 57 non-small cell lung cancer tissues and human non-small cell lung cancer cell lines was significantly up-regulated(P<0.05).The expression of Linc00842 was significantly down-regulated in cancer tissues and human non-small cell lung cancer cell lines.(P<0.05)3.The results of correlation analysis suggested that the high expression of Linc00665 in cancer tissues is associated with long-term living area and tumor pathological type.It is highly expressed in patients with lung adenocarcinoma and lung cancer in Xuanwei area,but low in lung squamous cell carcinoma and other pathological types and lung cancer in non-Xuanwei area and the difference was statistically significant(P<0.05).High expression of Linc00320 in cancer tissues was associated with lymph node metastasis,high expression in lymph node metastasis positive patients,and low expression in lymph node metastasis negative patients(P<0.05);Linc00324 high expression and clinical pathology of patients There is no obvious association of features.4.Linc00665,Linc00624,Linc00320,Linc00871,Linc00343,Linc003507 were up-regulated in A549,NCI-H1299,H1650,and HCC-827 compared to BEAS-2B,while Linc00842,Linc00578,Linc00544 showed the opposite result.Conclusion1.Linc00665,Linc00624,Linc00320,Linc00842 may be related to the development of non-small cell lung cancer2.Linc00665 and Linc00320 may become new biomarkers related to the diagnosis and treatment of non-small cell lung cancer,and Linc00665 provides new ideas for the further study of non-small cell lung cancer in the family of lung cancer in Xuanwei area.
