Research On Quality Evaluation Of Gentian In Jilin Province

Objective:To determine the contents of gentian polysaccharide,gentiopicroside,sweroside and swertiamarin in Gentian from different areas of Jilin Province.By analyzing and comparing the content difference.To provide evidence for the utilization and exploitation of Gentian.Determination of heavy metals of Pb and Hg and Pesticide residue in gentian from different areas.Provide a theoretical basis for its quality evaluation.Determination of gentian polysaccharide,gentiopicroside,sweroside and swertiamarin in gentian on the ground.To examine the medicinal properties on the ground.Analysis of gentiana from different producing areas in Jilin Province by HPLC,combined with relevant statistical method and establish a liquid chromatographic fingerprint.Provide scientific reference for the quality evaluation of gentian from different areas in Jilin Province.Method: 1.The experiment used water extraction and alcohol precipitation to extract gentian polysaccharide,And using UV to determine the content of gentian polysaccharide.2.Determination of gentiopicroside、swertiamarin and sweroside by HPLC;Determination of sweroside and swertiamarin in gentian by HPLC.3.HPLC gradient elution technique was used to analyze the active ingredients in gentian,and establish fingerprints and cluster analysis.4.Determination of Hg in gentian from different areas by AFC,Determination of Pb in gentian from different areas by FAAS.5.Determination of 7 organochlorine pesticide residues in gentiana by GC-ECD.Result:The method of anthrone method of inspection in the recovery rate was 99.67%,RSD value is 0.57%,polysaccharide sample solution absorbance of glucose solution and content within the range of 0.02~0.20mg(r=0.9995)showed a good linear relationship.Gentian polysaccharide content is 0.29%~1.13%.The content of gentiopicroside in 0.4~2.4μg(r=0.9999)has good linear relationship,Gentiopicroside content from 2.90% to 5.20%.The average recovery rate was 98.43%,RSD values were 0.76%.Sweroside showed a good linear relationship from 0.1~0.35μg(r=0.9999),and Swertiamarin showed a good linear relationship from 0.19~0.68μg(r=0.9999).The average recovery rate was 96.63%,96.91% and RSD values were 1.66%,1.49%.The content of Hg in gentian is between 20.3503 and40.3758ng/g;The content of Pb is between 147.9701 ~ 651.1752ng/g;The residues of pesticides in gentian from different areas of origin were lower than the relevant standards of the “Chinese Pharmacopoeia”(2015 edition)Conclusion: 1.There are some differences in the content of gentian polysaccharides in gentian from different areas.The high content of gentian polysaccharides in gentian stems and leaves,has a certain development value;2.Jilin Province gentian are either wild or cultivated,its gentiopicroside content meets the standards set by the “Chinese Pharmacopoeia”,The content of gentiopicroside in roots and rhizomes of gentian from different areas has a certain relationship with its ecological environment;3.The content of swertiamarin in gentian was higher,while the content of sweroside was lower.4.The results of heavy metal determination not only provide accurate reference for the limits of Hg and Pb in gentian medicinal materials,but also provide information for the production of authentic medicinal materials for gentian.And provide a reference for the quality specifications of the gentian medicinal herbs market;The pesticide residues in BHC、DDT、PCNB in gentian from different areas of origin,except Jiangmifeng town of Jilin Province,few were detected in gentiana from other regions of Jilin Province,that Jilin Province has better natural conditions.
