A Rapid Analysis Of Simultaneous Quantifcation Of 61 Drugs Of Abuse In Urine By UPLC-MS/MS

Objective:A rapid and sensitive ultraperformance liquid chromatography coupled with mass spectrometry(UPLC-MS/MS)method has been developed fo r simultaneous determination of 61 drugs of abuse(heroin、morphine、03-mon oacetylmorphine、06-monoacetylmorphine、oxycodone、hydrocodone、hydromor phone、oxymorphone、thebaine、cocaine、benzoylecgonine、methadone、EDDP、codeine、6-acetylcodeine、dihydrocodeine、dihydroetorphine、buprenorphine、nor buprenorphine、fentanyl、pethidine、caffeine、phenacetin、amphetamine、MA、MDA、MDMA、MBDB、MDEA、PMMA、ephedrine、N-methylephedrine、ket amine、norketamine、methcathinone、phencyclidine、mescaline、LSD、diazepam、nitrazepam、flurazepam、bromazepam、clonazepam、flunitrazpan、oxazepam、pr azepam、temazepam、7-aminontrazepam、7-aminofluninazepam、triazolam、alpro zolam、midazdam、estazolam、a-hydroxytriazolam、a-hydroyalprazolam、a-hydro xymidazolam、zopiclone、zolpidem tartrate、zaleplon、methaqualone、fenflurami ne)in urine.Methods:According to the ratio of 1:9(V:V),a mixture of urine sample and ultrapure water,a mixture of urine sample and methanol,a mixture of urine sample and acetonitrile and urine sample dealt with HLB,were respectiv ely vortexed in 30s and centrifuged with 8500r/min in 10 min,the supernatant was analyzed with UPLC-MS/MS system coupled with an electrospray ionizati on(ESI)source.The separations were carried out on an ACQUITY UPLC(?)H SS C18(2.1mm×50mm,1.8μm)at 50℃ and with a constant flow rate of 0.4 mL/min and the gradient-elution consisting of 0.1%formic acid in acetonitrile and a mixture of 5 mM ammonium formate in ultrapure water with pH3.The MS parameters were set as capillary voltage(3.0 kV),desolvation gas temper ature(500℃),desolvation gas flow(800 L/Hr)and collision gas flow(0.25 m L/min).The samples were measured in the mode of electraspray positive ioniza tion(ESI+)and multiple reaction monitoring(MRM).The target compounds w ere qualified by multiple characteristic ion pairs and quantified by the external standard calibration curve method.Results:Considering all these factors,the pretreatment of methanol is better than t he rest.Conclusion(s):Dealt with methanol and analyzed with UPLC-MS/MS,the method presented here is validated to be simple,rapid,sensitive and accurate for the determination of trace medicines and metabolites in urine samples.
