Ethanol ultrasonic extraction and supercritical CO2 extraction were taken to optimize extraction process of chlorogenic acid of wormwood and then used poloxamer-188 material to modify chlorogenic acid,improve its water solubility,to exert its bioavailability.(1)Ethanol ultrasonic,three kinds of factors which were ethanol concentration,solid-liquid ratio,ultrasonic time selected.The results of orthogonal test showed that the optimum conditions for extracting chlorogenic acid by ethanol ultrasonic method for the concentration of ethanol was 70%,liquid-to-solid ratio was 1:10,the ultrasonic time was 50 min,the chlorogenic acid extraction yield can reach 1.3%.(2)Supercritical CO2 extraction,according to single experimental results,we selected extraction temperature,pressure,flow rate of cosolvent to design optimization of the extraction process of response surface.The results of response surface optimization test showed that the optimum extraction process for chlorogenic acid extraction temperature was 50℃,extraction pressure was 20MPa,cosolvent velocity was 0.8 ml/min,the optimal chlorogenic acid extraction yield is1.42%,The optimization method is feasible and is better than ethanol ultrasonic method.We used poloxamer-188 material on extraction of chlorogenic acid coating modification,improve its water solubility,to exert its bioavailability,SEM results showed that chlorogenic acid has been successfully modified by poloxamer188,the HPLC results showed that chlorogenic acid’s water-solubility has been greatly improved after poloxamer-188 modified.(1)CFU and MIC,the results of CFU experiments showed that the bactericidal activity of poloxamer-188 chlorogenic acid was higher than that of chlorogenic acid.Experimental results showed that the MIC of S.aureus and E.coli were lower than chlorogenic acid.(2)Antioxidant experiment,the inclusions of,the experimental results show that the chlorogenic acid inclusion poloxamer-188,the scavenging rate of DPPH,·OH and O2-significantly lower than that chlorogenic acid group and the control Vc group.(3)Cell experiment,to study the effect of the encapsulation on cancer cells,the LIVE/DEAD experiment results showed that poloxamer-188 chlorogenic acid has a significant inhibitory effect on the activity of gastric.(4)Fruit preservation experiment,the pear was treated for 14 days and the weight loss rate,total sugar content,and Vc content were measured on a regular basis.The experimental results showed that the weight loss rate of Pyrus pallioxam-188 inclusion complex chlorogenic acid group was obviously lower than the blank control group and chlorogenic acid group;the total sugar and Vc content were always higher than the other two groups. |
