Background:Background: With the deterioration of the environment and the progress of science and technology,more and more lung tumor patients need treatment.These patients require surgical treatment and subsequent chemoradiotherapy.The probability of fungal infection increased significantly after surgery or after chemotherapy.Candida albicans is a two-state fungus as part of a normal human microbiome.Candida albicans are symbiotic organisms that do not usually cause infection.In recent years,the infection of candida albicans has increased by more than 20 times due to the widespread use of immunosuppressant,broad-spectrum antibiotics and anti-tumor drugs.Candida albicans has become the fourth most common bacterial flora in the hospital.Candidiasis is more likely to occur when the skin,mucosal mechanical barrier is damaged or the body's immune system is compromised.The occurrence and development of candida albicans infection depends on three aspects: the number of candida albicans,the host's immune function,and the way the fungus invades the organism under different environmental conditions.Candidiasis is more likely to occur when the skin,mucosal barrier is damaged or the body's immune system is compromised.Because many patients with thoracic surgery are malignant tumor patients,after thoracotomy,the patient's skin mucosal barrier is damaged,the body immunity is relatively low,and the probability of pulmonary fungus infection is greatly increased.There is no obvious specificity in the clinical manifestations or symptoms of a fungal infection,especially after a deep fungal infection.The diagnosis of candida albicans infection is difficult,and there is no simple and effective diagnostic method or method.The above characteristics cause the misdiagnosis and missed diagnosis of candida albicans infection.At the same time,fungal infection is not easy to control,and if it cannot be treated timely and effectively,it often leads to poor prognosis and high fatality rate.MP,as the outermost layer of the cell wall of candida albicans,is the first site for fungal and host immune response.It plays an important role in candida infection.MP is not only necessary to maintain cell shape and stability,but also with candida immunogenicity and virulence.Although a lot of research has been done,the detailed role of MP in the pathogenesis of candida albicans is not fully understood.Mannose glycoprotein dialogue candida cell wall structure at home and abroad and its pathogenic mechanism,and immunology research is more,this topic in the use of the cell walls of c.albicans mannose glycoprotein stimulate macrophage RAW264.7 mice after macrophages from general characteristics,the release of cytokines,and signaling pathways that may be relevant aspects for the following research.Methods: The macrophages of mouse macrophages were stimulated and cultured with different concentrations of MPs(50,100,200,g/ml)and LPS;1.The proliferation and activity of cells were detected by MTS reagents;2.The phagocytic ability was observed by macrophage phagocytic phagocytes,and the morphological changes of the cells were observed under the microscope;3.The cell apoptosis was detected by flow cytometry;4.RT-PCR was used to detect JAK2,STAT3,IL-1,IL-6 and TNF-alpha in RAW264.7cells;5.ELISA method was used to detect the secretion of il-1,il-6 and TNF-in culture;6.Protein expression of JAK2,p-JAK2,STAT3 and p-STAT3 was detected by western blot.Results: 1.MP can promote the proliferation of RAW264.7 cells and enhance their cell activity;2.MP inhibits its phagocytic ability;3.MP has no significant effect on apoptosis of macrophages,but can promote the apoptosis of macrophages induced by LPS.4.MP promotes the release of macrophage cytokines(IL-1,IL-6,TNF-a),while promoting the release of lps-induced cytokines;5.MP can be up-regulated by p-JAK2,STAT3 and p-STAT3,with no significant effect on JAK2 expression.Conclusion: MP can promote the occurrence of LPS induced macrophage inflammation and participate in the inflammatory immune process of organisms.By activating the JAK2/STAT3 signaling pathway,the transformation of the macrophage RAW264.7 to the M1 type was promoted to promote the occurrence of inflammation.
