Objective: To analyse the expression of Isocitrate dehydrogenase 1(IDH1)gene mutation and O6-methylguanine-DNA methyltransferase(MGMT)promoter in different glioma grades.To discuss the correlation between IDH1 gene mutations and MGMT promoter methylation.To study their role in the development of glioma disease and provide valuable help for the diagnosis,individualized treatment and prognosis of glioma.Methods:Thirty two pathology confirmed cases of glioma specimen by surgical resection in Shengjing Hospital affiliated to China Medical University were collected,which were sent to the Genetron Health company for molecular testing.IDH1 gene mutation detection using QPCR-Taqman and Sanger method,MGMT promoter methylation detection using pyrosequencing.Spearman correlation analysis was performed on the correlation between IDH1 gene mutation and MGMT promoter methylation in patients with glioma.Results: In 32 glioma patients,there are ten cases of IDH1 gene mutation positive expression,the total positive rate was 31.3%.The positive expression rates of IDH1 gene mutation in WHO ?,? and ? gliomas were 72.7% and 15.4%,0.0% respectively.There was statistically significant among them(P <0.05).MGMT promoter methylation in 17 cases,the total methylation positive rate was 53.1%,MGMT promoter methylation positive expression rate was no significant difference in different glioma grades(P>0.05).No correlation was found between IDH1 gene mutation and MGMT promoter methylation(P> 0.05).Conclusion: The positive rate of IDH1 gene mutation in different grades of glioma is different,with the pathology level increased its positive expression rate decreased.The positive rate of MGMT promoter methylation was lower in WHO ? grade and there is no significant among the three grades.There is no correlation between IDH1 mutation and MGMT promoter methylation.
