Part I Changes of vertebral bone mineral density and relationship between vertebral bone mineral density and serum GH,IGF1,IGFBP3,BMP2,BMP9,E2 in adolescent idiopathic scoliosisObjective:To investigate Changes of vertebral bone mineral density and relationship between vertebral bone mineral density and serum GH,IGF1,IGFBP3,BMP2,BMP9,E2 in adolescent idiopathic scoliosis.Methods:118 patients with idiopathic scoliosis(aged from 10~16 years,41 males and 77 females,Cobb’s angle 10°~60°)and 125 age-matched healthy subjects(aged from 10~16 years,43 males and 82 females)as control.The patients were divided into S type scoliosis group and C type scoliosis group according to the direction of the scoliosis.According to the Cobb’s angel divided the patients into low risk group(10°~20°),mid risk group(20°~ 40°),high risk group(> 40°).The bone mineral density of scoliosis vertebral body of patients and the corresponding vertebral body of control groups were detected by CT.The serum GH,IGF1,IGFBP3 and E2 levels of all subjects were determined by using Chemiluminescent Immunoassay(CLIA).The serum BMP2 and BMP9 were determined by Enzyme Linked Immunosorbent Assay(ELISA).Results:The vertebral bone mineral density,serum IGF1,IGFBP3,BMP9 levels of S type scoliosis group and C type scoliosis group decreased significantly by comparing with the control group(p<0.05).The difference of serum GH,BMP2,E2 level between patients and the control group was not statistically significant.The serum E2 levels of C type scoliosis group increased significantly by comparing with the S type scoliosis group.The level of IGF1 and IGFBP3 was positively correlated with bone mineral density(r=0.517,p<0.05、r=0.678,p<0.001).The serum GH levels of mid risk group,high risk group with C type scoliosis were decreased significantly by comparing with the low risk group(p<0.05).The serum E2 levels of high group with C type scoliosis were decreased significantly by comparing with the low risk group and mid risk group(p<0.05).Conclusion:There was a lower vertebral bone mineral density in patients with adolescent idiopathic scoliosis,and the decreased expression of IGF1,IGFBP3,BMP9 may be one of the important reasons for the reduction of bone density.The bone development of S type scoliosis group and C type scoliosis are different.Part II A pilot study on serum GH,IGF1,IGFBP3,BMPs in Congenital ScoliosisObjective:To investigate the serum levels of GH,IGF1,IGFBP and BMPs in Congenital Scoliosis.Methods:37 patients with Congenital Scoliosis(from January 2015 to August 2017,aged from 1~16 years,15 males and 22 females,Cobb’s angle 10°~75°)and 46 age-matched healthy subjects(from January 2015 to August 2017,aged from 1~16 years,20 males and 26 females)as control.The serum GH,IGF1,IGFBP3 and E2 levels of all subjects were determined by using Chemiluminescent Immunoassay(CLIA).The serum BMP2 and BMP9 were determined by Enzyme Linked Immunosorbent Assay(ELISA).Detection of BMP2,4,6,7,9 expression levels of leukocytes in blood by Real Time PCR method.Results:The levels of serum IGF1,IGFBP3 and BMP9 and the level of BMP9 expression in blood leukocytes of CS children were significantly lower than those in the control group(p<0.05).The levels of serum GH,BMP2,BMP4,BMP6 and BMP7 were not significantly different from those of the control group.Conclusion:The serum IGF1,IGFBP3 and BMP9 levels of patients with Congenital Scoliosis were lower than control,which may be one of the important factors of scoliosis.
