| Background:Cataracts are the most common cause of vision loss in people over age 40 and is the principal cause of blindness in the world.Although in this modern century the cataract surgery had been improve so much,but there is still some complication of after cataract surgery,therefore we did this experiment,we tried to treat the cataract without the cataract surgery.Objective:To analyze the effect of Lanosterol in pig lens cataractogenesis and analyze which cataractogenesis methods cause the worst cataract imitation in pig lens.Methods:We used fresh pig’s eyeball that obtained immediately after sacrifice at local slaughterhouse,the sacrificed pigs are 6 months old.We induce the sacrificed pig’s eye lens to mimic the cataract in human eye lens.We induce the cataract in pig’s eye lens by 6 methods of cataractogenesis,PBS injection to the anterior chamber of pig’s eyeball,fixative agent injection to the anterior chamber of pig,s eyeball through the limbus of pig’s eyeball(Formalin:Isopropyl Alcohol:Ethanol in a 4:3:3 ratio),fixative agent injection to the vitreous body of the pig’s eyeball,immersion of the dissected pig’s eye lens in fixative agent,immersion of the dissected pig’s eye lens in PBS,stored pig’s eyeball in 4℃ freezer for 24 hours.Note that all of the 5 methods of cataractogenesis pig’s lens was stored in room temperature 25.8℃ for 24 hours,except the 4℃ freezer cataractogenesis.Every cataractogenesis methods,we use 5 pig’s eye lenses in every group.Every cataractogenesis pig’s lens was then photographed and we count the light absorbance with spectrophotometry.We prepared the mixture of lanosterol and then stored the lanosterol mixture in room temperature and no light condition place.The treatment of the cataractogenesis pig’s eye lens with lanosterol,the we count the light absorbance,the transmittance and photographed the cataractogenesis pig’s eye lens before treatment with lanosterol,without treatment of lanosterol,and after treatment with the mixture of lanosterol,the treatment is the immersion of the cataractogenesis pig’s eye lens in the mixture of lanosterol for 24 hours in room temperature 25.8℃.Results:We only choose the 2 methods of cataractogenesis pig’s eye lens which is immersion of cataractogenesis pig’s eye lens in fixative agent(Formalin:Isopropyl Alcohol:Ethanol in a 4:3:3 ratio)for 24 hours in room temperature 25.80C,and the stored pig’s eyeball in 40C freezer cataractogenesis.Because only the two of the cataractogenesis pathway has the worst light absorbance and the transmittance.After the treatment with the mixture of lanosterol,both the immersion of pig’s eye lens in fixative agent cataractogenesis and the stored pig’s eyeball in 4℃ freezer cataractogenesis are seems worse than the before treatment with the mixture of lanosterol.The photographed both the immersion of cataractogenesis pig’s lens in fixative agent and the stored pig’s eyeball in 4℃ freezer cataractogenesis also shows the after treatment of both cataractogenesis lens seems even worse than before treatment of the cataractogenesis lens.Conclusion:the reversed protein aggregation in natural cataract with lanosterol in rabbit lens and dog’s eyeball seems does not work with cataract induction using pig’s eye.Maybe the fixative agent(Formalin:Isopropyl Alcohol:Ethanol in a 4:3:3 ratio)cataractogenesis with pig’s eyeball are too complicated to be reversed its protein aggregation in cataract,maybe we should used the naturally occurring cataract in animal. |
