| ObjectiveLung cancer has been increasing both in incidence and mortality in China.Long noncoding RNAs(lncRNAs)controls almost all biological processes.LncRNA HOX transcriptional antisense RNA(HOTAIR)inhibits gene expression through its related proteins and increases the proliferation,invasion,metastasis and drug resistance of lung cancer cells.High expression in lung cancer is associated with lung cancer metastasis and poor prognosis.The purpose of this study was to investigate SNP polymorphism of HOTAIR at four sites of RS920778(C> T),RS12826786(C> T),RS4759314(A> G)and RS1899663 Relationship of genetic susceptibility to lung cancer.MethodsThis study selected 262 patients with primary lung cancer and 451 healthy controls as subjects,all subjects were drawn 5ml venous blood.The high-throughput Taqman-MGB probe technique was used to genotype RS920778(C> T),RS12826786(C> T),RS4759314(A> G)and RS1899663(G> T).The primary lung cancer patients were divided into several subgroups including age,gender,smoking history and pathological types of patients with different clinical features.Non-conditional logistic regression analysis was applied in HOTAIR single nucleotide polymorphisms and primary lung cancer risk correlation.We employ SPSS18.0 and SHESIS software for statistical analysis of data,and each loci for linkage disequilibrium and haplotype analysis.Different survival rates of different genotypic patients with lung cancer were analyzed through Kaplan-Meier.ResaltsIn our study,RS920778(C> T)and RS1899663(G> T)were highly correlated with susceptibility to primary lung cancer among the four SNPs.In the primary lung cancer group,RS920778(The frequency of C / T genotype(C / T + TT)was higher than that of control group(OR = 1.467,95% CI: 1.069-2.014,P <0.05).The TT genotype frequency of RS1899663(G> T)locus was significantly higher than that of wild-type GG genotype(OR = 3.220,95% CI: 1.272-8.050,P <0.05).There was no significant correlation between RS12826786(C> T)and RS4759314(A> G)and the incidence of primary lung cancer.Subsequent studies indicated that the C / T genotype(C / T + TT)at RS920778(C> T)loci was significantly lower in all subgroups stratified by age,sex,smoking history and pathological type)Was significantly associated with susceptibility to male,smokers,and primary lung cancer patients whose pathological type was squamous cell carcinoma.A subsequent linkage study between single nucleotide polymorphism in HOTAIR and genetic predisposition to primary lung cancer and haplotype analysis revealed that linkage disequilibrium and haplotype analysis detected HOTAIR gene polymorphisms and The risk of lung cancer.RS920778(C> T)and RS1899663 sites,there is a highly linkage disequilibrium(D ’= 0.86,r2 = 0.52).ConclusionsOur results show that HOTAIR gene RS920778(C> T)and RS1899663(G> T)single nucleotide polymorphisms are associated with genetic predisposition to primary lung cancer in Chinese population and may increase the risk of developing primary The risk of lung cancer.The increased susceptibility to RS920778(C> T)C / T genotype(C / T + TT)was associated with gender,smoking history,and pathologic typing.This study revealeded that HOTAIR SNP was associated with increased susceptibility to lung cancer Perceptual play an important role.In the future for its in-depth study of the mechanism is necessary... |
