Object.Cytomegalovirus(CMV)infection is a serious complication of liver transplantation.In this study,we investigated the change of T-helper 1(Th1)type cytokine interferon gamma(IFN-?)mRNA,Th2 type cytokine interleukin 10(IL-10)mRNA,and Th3 type cytokine transforming growth factor beta 1(TGF-?1)mRNA expression in peripheral blood mononuclear cells(PBMCs)in infants living related liver transplantation recipients with CMV infection.Methods.A total of 40 children with biliary atresia who underwent living related liver transplantation were collected from 2016 to 2017 at Tianjin First Central Hospital organ transplantation center.The 40 patients were divided into 20 patients with CMV infection and 20 patients of without CMV infection.Peripheral blood mononuclear cells(PBMCs)from the 40 patients were isolated from whole blood by Ficoll density gradient centrifugation using a commercially lymphocyte separation medium.The total RNA was extracted from PBMCs from the 40 patients,and then subjected to reverse transcription using a cDNA Reverse Transcription kit.The levels of Interferon gamma(IFN-?),interleukin 10(IL-10),transforming growth factor beta1(TGF-?1)mRNA were determinated using fluorescence quantitative reverse transcription polymerase chain reaction(qRT-PCR)analysis.The changes of the CD4/CD8 ratio of PBMCs from the 40 patients were determinated by flow cytometry.Result.The expression levels of IL-10 mRNA were significantly increased in the CMV infection group compared with those in healthy group(P<0.05).The expression levels of TGF-?1 mRNA were significantly increased in the CMV infection group compared with those in stable group or healthy group(P<0.01).However,the IFN-? mRNA expression level was significantly down-regulated in the CMV infection group compared with that in the stable group or the healthy group(P<0.01).The expression levels of IL-10 and TGF-?1 mRNA were not significantly difference in the stable group compared with the healthy group(P>0.05).The ratios of CD4/CD8 were significantly lower in the CMV infection group compared with those in stable group(P<0.05).After the administration of antiviral drugs such as ganciclovir to recipients with CMV infection,the expression level of IFN-? mRNAwas significantly increased compared with that at the time of CMV infection(P<0.05).Conclusion.Low level of Th1 type cytokine IFN-? mRNA seems to correlate with CMV infection in infants living related liver transplantation recipients.
