Objective:The effect of myocardial injury caused by macrophages in SAP is not yet clear.In this study,we investigate the protective effects of coldronate-liposome on myocardial injury by SAP.Method:48 Sprague-Dawley rats were selected into three groups(Sham group,SAP+BLANK group and SAP+LC group).Each group was subdivided into 2 h and 6 h.There were 8 rats in each group.The changes of IL-6,amylase and TNF-a in rats at 2 and 6 h were measured.The effects of LC on pathological changes of myocardial tissue in SAP rats were observed and compared.Macrophages between myocardium was observed in SAP rats by staining of prussian blue.The dynamic changes of Akt and MAPK expression in SAP rats were observed by immunohistochemistry.Result:The contents of IL-6,amylase and TNF-a were higher at 2,6 h in SAP+LC treatment group and SAP+BLANK group than sham group(P <0.05).The contents of IL-6,amylase and TNF-a in SAP+LC treatment group were significantly lower than those in SAP+BLANK group(P <0.05).Compared with Sham group,the histological score at 2,6 h was significantly increased in SAP+BLANK group(P <0.05).The histological score at 2,6 h in the SAP+LC group was significantly lower than SAP+BLANK group.(P <0.05).The number of macrophages were increased in SAP+BLANK group compared with SAP+LC group at 2,6 h by prussian blue staining.Compared with Sham group,the Akt and MAPK score at 2,6 h in SAP + BLANK group was significantly increased(P <0.05).The Akt and MAPK score at 2,6 h in SAP + BLANK group was significantly increased than SAP+LC group.(P <0.05).Conclusions:LC can decrease the content of IL-6,amylase and TNF-a,improve the pathological changes of myocardial injury during SAP and selectively remove monocytes/macrophages.LC can reduce the expression of Akt and MAPK in myocardium.LC can reduce pancreatic injury and myocardial injury caused by SAP in rats.
