Objective:This study detected the expression of microRNA-135b?miR-135b?and microRNA-141?miR-141?in cancer tissues and the corresponding upper marginal no tumor mucosal tissue of colorectal cancer patients,and to analyze their relationship with the clinicopathological characteristics of patients.And to investigate the value of miR-135b and mi R-141 expression in early warning?diagnosis and treatment in colorectal cancer.Method:1.Collected 28 fresh cancer tissues of patients with colorectal cancer surgery in anorectal surgery and tumor surgery of Tangshan workers'hospital from January 2015 to January 2016.28 cases of corresponding upper marginal mucosal tissues were selected and put into the liquid nitrogen after centrifuged,then Stored in-80?refrigerator.2.Extracted total RNA of the tissues and transcribed into c-DNA after verified the integrity of it.Measure the expression of miR-135b and miR-141in the tumor tissues and their corresponding upper marginal mucosal tissues through the method of real-time quantitative PCR.3.Analyzed the relationship between the relative expression of mi R-135b and miR-141 and gender,age,position,infiltrating degree,differentiation degree,and tumor markers?CEA,CA199,CA724?in patients.4.SPSS19.0 software was used for statistical analysis,all the data was represented as median?interquartile range?,Wilcoxon test was used to analyze the data between two groups and<0.05 represents statistical significance.Result:1.ression of miR-141 in colorectal cancer tissues28 samples compared with Ct value of the upper marginal mucosal tissues.To calculate the relative expression of miR-141 in colorectal cancer tissues and the upper marginal no tumor mucosal tissues by 2-??Ct method.The results showed,the relative expression of miR-141 in the upper incised margin and colorectal cancer tissues were 1.581?0.822,4.278?0.393?0.286,1.148??P=0.001?.Compared with the upper marginal no tumor mucosal tissues,the expression of mi R-141 was significantly reduced in colorectal cancer tissues,the difference was statistically significant.2.Analysis of the relationship between mi R-141 and clinicopathologic features of colorectal cancerCalculated the relative expression of mi R-141 in colorectal cancer tissues and the upper marginal no tumor mucosal tissues by 2-??Ct method.The mean value of colorectal cancer tissues of each sample was divided by the mean value of the upper marginal no tumor mucosal tissues,the result was the relative expression of miR-141.The results showed,The relative expression of colorectal cancer patients with no lymph node metastasis and lymphatic metastasis were 1.002?0.424,1.577?20.350?0.265,0.861??P=0.011?.The relative expression of patients with CA199<35U/ml and?35U/ml were1.441?1.054,4.780??0.364?0.269,0.899??P=0.004?.The relative expression of colorectal cancer patients with CA724<6.9U/ml and?6.9U/ml were1.452?1.065,4.860?0.384?0.281,0.957??P=0.034?.3.Expression of miR-135b in colorectal cancer tissues28 samples compared with Ct value of the upper marginal mucosal tissues.To calculate the relative expression of miR-135b in the upper marginal no tumor mucosal tissues and colorectal cancer tissues by 2-??Ct??Ct method.Result showed,the relative expression of miR-135b in the upper incised margin and colorectal cancer tissues were 1.222?0.800,1.455?and1.732?0.957,2.222??P=0.019?.Compared with the upper marginal no tumor mucosal tissues,the expression of miR-135b was significantly increased in colorectal cancer tissues.4.Analysis of the relationship between miR-135b and clinicopathologic features of colorectal cancerCalculated the relative expression of miR-135b in colorectal cancer tissues and the upper marginal no tumor mucosal tissues by 2-??Ct method.The mean value of colorectal cancer tissues of each sample was divided by the mean value of the upper marginal no tumor mucosal tissues,the result was the relative expression of mi R-135b.The results showed,The relative expression of colorectal cancer patients with no lymph node metastasis and lymphatic metastasis were 1.426?0.748,1.964?and 2.034?1.728,3.430??P=0.025?.The relative expression of patients with CA199<35U/ml and?35U/ml were 1.448?0.796,1.959?3.635?2.265,11.374??P=0.002?.The relati ve expression of colorectal cancer patients with CA724<6.9U/ml and?6.9U/ml were 1.652?0.865,1.999?3.694?2.273,11.596??P=0.019?.Conclusion:1.The low expression of mi R-141 in the tissues of colorectal cancer may be related to the invasion of colorectal cancer.2.The high expression of miR-135b in the tissues of colorectal cancer may have a certain correlation with the occurrence of malignant invasive biological behavior of colorectal cancer.
