Object:Our study aims to establish a mouse mode of iron overload by intraperitoneal injection of iron sucrose injection and investigate the impcat of iron overload on poor graft function after hematopoietic stem cell transplantation.Methods:1.A total of 35 BALB/C mice were divided into control,lower dose(5mg/ml),middle does(10mg/ml)and high dose(20mg/ml)group.the group of lower dose,middle does and high dose group were injectde with iron sucrose injection(0.2ml)three times a week for four weeks.Iron overload were confirmed by observing the bone marrow and hepatic and splenic deposits,the level of serum ferritin and organ coefficient of liver and spleen.2.A total of 42 BALB/C mice were divided into control,iron,health irradiation,health transplantation,iron irradiation,iron transplantation groups.And After four weeks,the group of health irradiation,health transplantation,iron irradiation,iron transplantation groups accept a total of 8 Gy X-ray,next day,the normal mice bone marrow cells of donor C57BL/6 mice transfused into the iron transplantation and ihealth transplantation groups mice.After 2 weeks,hematopoietic recontruction were confirmed by periphheral blood.In addition,bone marrow deposits and colony forming activity were monitored.Result:1.The iron overloaded mouse model was successfully established,Iron deposits in bone marrow,liver,and spleeen were markly increease in mouse modles,compared with control,the liver coefficient [6.17±0.79 VS 5.06±0.46] of iron group,iron irradiation group,iron transplantation group was increasesignificiantly(P<0.05),the splen coefficient [0.62±0.12 VS 0.42±0.09]of iron grou,iron irradiation group,iron transplantation group was increase significiantly(P<0.02).2.Iron overload damage bone marrow hematopoietic function.include:(1)Compared with health transplantation groups,the iron transplantation group the amounts of red blood cells were decreased significiantly [(6.42±1.00)×10^12/L Vs(9.93±2.67)×10^12/L],P<0.001,hemoglobin were decreased significiantly[95.85±14.55 g/L Vs 116.00±28.01g/L],P<0.001;(2)The iron transplant ation group bone marrow cellularity was significiantil lower compared with irradiation+transplantation group;(3)The colony forming activity of iron transplantation was lower compared with health transplantation group,the number of granulocyte macrophage colony forming unitwas lower significiantly [24.90±2.60 Vs 48.50±1.00],P<0.002,the number of erythroid colony forming unit was lower significiantly[20.20±2.30 VS 36.40±5.00],P<0.005,the number of mixed colony forming unit was lower significiantly[6.70±3.20 VS 27.30±1.70],P<0.001.3.Compaired with control group,the hemoglobin of iron group was increase significiantly [(135.85±16.84)g/L VS(111.14±16.06)g/L],P<0.001,and the amount of plataletwas increase significiantly [(949.14±199.31)×10^9/L Vs(649.57±280.44)×10^9/L ],P<0.001;Conclusion:1.The iron overloaded mouse model was successfully established by intraperitoneal iron suorose injection;2.Iron overload could damage bone marrow hematopoietic function aggravatingly,which may lead to poor graft functions after hematoietic stem cell transplantation. |
