【Objective】 The neuroprotective effect of rehabilitation training after cerebral ischemia has been the consensus of scholars at home and abroad.However,there are many mechanisms for it and it is not yet completely clear.In this study,the middle cerebral artery occlusion(MCAO)model was established to perform rehabilitation training.The neurological recovery of rats was observed,and the proliferation of new neural stem cells was detected,and the related markers of neonatal neural stem cells were measured.To explore the mechanism of rehabilitation training on neurological recovery and nerve regeneration in rats with cerebral ischemia.【method】 1.The rat model of MCAO cerebral ischemia was established by the method of suture.2.Longa 5-point system NDS method and modified neurobehavioral score(mNSS)were used to evaluate neurological function changes in each group.3.Using magnetic resonance to measure the volume of cerebral infarction in each group 4.On the 7th day after modeling,the subventricular zone of the lateral ventricle was taken and the brain tissue sections were prepared.Using laser confocal microscopy,DCSC and Brdu were used to label newborn neural stem cells.The number of newborn neural stem cells in each group was observed.Happening 5.After 7 days of modeling,the brain tissue near the subventricular zone of the lateral ventricle was taken and the activation of PI3K/Akt signaling pathway was detected by Western blot.6.PI3K/Akt signaling pathway inhibitor Ly294002 was injected into the lateral cerebral ventricle after modeling.The periventricular subventricular zone was collected on the 7th day after modeling.The expression of DCX and Nestin was detected by Western blot.【result】 1.Rehabilitation training can improve the neurological function of rats with cerebral ischemia 2.Rehabilitation can reduce infarct volume in rats with cerebral ischemia 3.Increased number of neonatal neural stem cells in subventricular zone of lateral ventricle in rats in MCAO+ rehabilitation training group 4.Rehabilitation can activate phosphorylation of PI3K/Akt signaling pathway in MCAO rats 5.Ly294002,a phosphorylation inhibitor of PI3K/Akt signaling pathway,inhibits regeneration-mediated expression of neonatal neural stem cell markers 【Conclusion】 Rehabilitation training has a protective effect on neurological function after cerebral ischemia,and its mechanism may be related to the promotion of regeneration of neonatal neural stem cells.
