| ObjectsCervical cancer is the most common malignant tumor in female genital tract,and its pathogenesis is not yet fully understood.Golgi phosphoprotein-3(Golpi phosphoprotein 3,GOLPH3)has been shown to participate in a wide variety of tumor diseases,its role in cervical cancer is remarkable.This paper intends to explore GOLPH3 in cervical squamous cell carcinoma,cervical squamous intraepithelial neoplasia and expression in normal cervical tissue,the analytical expression differences,combined with clinical information,understand the clinical significance of GOLPH3 abnormal expression,in order to development mechanism of the cervical squamous cell carcinomas and its diagnosis.MethodsCollected in January 2016-December 2016 in the second affiliated hospital of fujian medical university hospital of department of gynaecology patients 84 cases(see text)inclusion criteria: normal cervix group(for patients with uterine fibroids whole hysterectomy)42 cases,21 cases of cervical squamous epithelial dysplasia group,21 cases of cervical squamous carcinoma group,cervical tissue samples,using immunohistochemical,protein immunoblot and polymerase chain reaction method to detect the tissue GOLPH3 in cells,proteins and gene expression level,and analyze its role in the development of cervical squamous carcinoma.Results 1.Cervical squamous group,cervical squamous epithelial dysplasia and the number of normal cervical tissue pregnancy,marriage age,culture level,household registration and the abortion,there were no statistically significant difference(P > 0.05);Cervical squamous carcinoma group age,delivery times,hospitalization days,operation time and hospitalization expenses were significantly higher than that of cervical squamous epithelial dysplasia group and the control group,the difference was statistically significant(P < 0.05),but the cervical squamous epithelial dysplasia group delivery times,surgery,and surgery time were significantly lower than the control group,the difference was statistically significant(P < 0.05),and cervical squamous epithelial dysplasia group age,delivery times compared with control group,no statistically significant difference(P > 0.05).2.The expression of go lph 3 protein was detected in cervical squamous cell carcinoma group,cervical squamous dysplasia group and normal cervical tissue.the levels of histone in cervical squamous cell carcinoma group and cervical squamous epithelial dysplasia group were significantly higher than those in normal cervical tissue(p < 0.05).the expression level of go lph 3 protein in cervical squamous cell carcinoma group was higher than that in cervical squamous epithelial dysplasia group(p < 0.05).3.The expression of go lph 3 mrna was detected in cervical squamous cell carcinoma group,cervical squamous dysplasia group and normal cervical tissue.the mrna levels of cervical squamous cell carcinoma group and cervical squamous dysplasia group were significantly higher than that of normal cervical tissue(p < 0.05).the expression of go lph 3 mrna in cervical squamous cell carcinoma group was higher than that in cervical squamous dysplasia group(p < 0.05).4.In cervical cancer patients,with the increase of clinical staging,enhance Golph 3 expression,which can increase the length of hospital stay,operation time,hospitalization expenses,and its not subjected to patient’s age,number of pregnancy and biochemical indexes such as the impact of individual differences.Conclusion 1.Cervical tissue Golph 3 expression quantity change,may promote the pathological process of cervical cancer,and to play a role in the occurrence of cervical cancer development,and to a certain extent can be as an indicator to predict the prognosis of cervical cancer.2.Cervical cancer incidence tends to be younger,which is closely related to age,preterm birth and HPV infection,and significantly increases with the number of days of hospitalization,hospitalization expenses and hospital stay. |
