Expression Of Claudin-3 In Colorectal Carcinoma And Its Clinicopathological Significance

Objective:To investigate the expression of claudin-3 in colorectal cancer patients,colorectal adenomas patients and the colon or rectum mucosa of normal individuals,and to explore the relationship between claudin-3 and the occurrence,invasion,and metastasis of colorectal cancer.The correlation provides a potential target for early detection and clinical treatment of colorectal cancer.Methods:A total of 82 patients who underwent surgery or endoscopic treatment at our hospital between September 2016 and June 2017 were collected.Among them,32 patients underwent surgical resection of colorectal cancer and 25 patients underwent colorectal adenoma surgery.Microscopic resection included endoscopic high-frequency resection,endoscopic mucosal resection(EMR)or endoscopic mucosal dissection(ESD)and colon or rectal biopsy of 25 normal subjects.Routine HE staining was used to observe the morphological changes under microscope in each group of specimens to distinguish the pathological features of histological grade and TNM staging of colorectal cancer.The expression of claudin-3 was detected by Western blot.Rerults: The expression level of claudin-3 in colorectal cancer tissues(0.91589±0.69015)was significantly higher than that of colorectal adenoma tissues(0.47748±0.33780)and normal tissues(0.203159±0.21827).The difference was statistically significant(t=3.123,t= 5.459,P<0.05),Spearman correlation analysis was moderately positive(r=0.490,P=0.000).The lymph node metastasis group(1.29604±0.49018)was significantly higher than the no lymph node metastasis group(0.48504±0.65092),and the difference was statistically significant(t=4.010,P<0.05).The correlation analysis showed a moderate positive correlation(r=0.675,P=0.000).In the TNM stage,the expression level of claudin-3 in stage III colorectal cancer tissue(1.24177±0.45043)was significantly higher than that in stage I colorectal cancer tissue(0.22594±0.34202)and stage II colorectal cancer tissue(0.57926±0.72213).The difference was statistically significant(t=4.185,t=2.943,P<0.05),and Spearman correlation analysis showed that there was a high positive correlation between the expression level of claudin-3 and TNM staging(r=0.709,P=0.000).The level of expression in the well-differentiated(grade 1)group(0.13552±0.17606)was lower than that in the moderately differentiated(grade 2)group(0.88125±0.67663),with a statistically significant difference(t=2.106,P<0.05);The expression level in the moderately differentiated(grade 2)group(0.88125±0.67663)was also lower than that in the poorly differentiated group(1.48591±0.37522),and the difference was statistically significant(t=2.362,P<0.05).The Spearman correlation analysis showed that there was a moderate positive correlation between the expression level of claudin-3 and the degree of differentiation(r=0.650,P=0.000).It has nothing to do with the patient's age,gender,tumor size and tumor position.Conclusions: Claudin-3 is closely related to the occurrence,differentiation and metastasis of colorectal cancer.However it has nothing to do with the patient's age,gender,tumor size and tumor position.Claudin-3 may be an indicator for the early diagnosis of colorectal cancer and may provide a potential therapeutic target for clinical treatment of colorectal cancer.
