To investigate the protective effect of the thymoquinone against radiation induced mortality,oxidative damage and small intestinal injury.A total of 110 male C57/BL6 mice(22-24g)were matched according to body weight and randomly assigned to one of three groups:control,total body irradiation exposure(IR,7.5Gy)only,and IR+Thymoquinone(IR+TQ)(10mg/kg).30 days survival rate were observed in the experiment.In oxidative damage experiment,we assayed the MDA level,the SOD level and the GSH level in liver and serum.In small intestinal injury experiment,we assayed the Lgr5,Ki67,Lysozyme,Villi,caspase8,caspase9,caspase3,Bax,p53 and yH2AX expression levels in the mice intestine crypts.We found that the 30 days survival rate were 100%(control),0(IR)and 30%(IR+TQ),respectively.Compared with the IR,the MDA level of the IR+TQ group reduced,the difference had statistically significant(p<0.05);Compared with the IR,the SOD level of the IR+TQ group increased,there are no statistical significance,but compared with the IR group,the SOD lever in serum increased,the difference had statistically significant(p<0.05);The GSH level of the IR+TQ group increased,the difference had statistically significant(p<0.01).Compared to the IR groups,the positive cells of Lgr5,Ki67 and Paneth in crypt cells increased 70.51%,47.96%and 32.69%,respectively;The positive cells of P53 decreased 52.24%.Significant difference was found between the IR group and the IR+TQ group(p<0.01).Thymoquinone potently protected against radiation-induced lethal,oxidative damage and small intestinal injury in mice,and it may be a potential radioprotector.
